白藜芦醇对新生大鼠软骨细胞增殖和Ⅱ型胶原表达的影响

摘　　要：[目的]探讨不同浓度的白藜芦醇对体外培养的新生大鼠软骨细胞增殖和Ⅱ型胶原表达的影响。[方法]不同浓度的白藜芦醇干预新生SD大鼠的软骨细胞24 h,分别观察不同浓度组细胞形态的变化、CCK-8方法检测白藜芦醇对软骨细胞增殖的影响和蛋白印迹分析法(Western blot)检测Ⅱ型胶原和PCNA的表达情况。[结果]CCK-8结果显示白藜芦醇对SD大鼠软骨细胞增殖有明显的抑制作用,且与剂量浓度呈正相关。与空白对照组和阳性对照药IL-1β相比,随着浓度的逐渐加大,Ⅱ型胶原和PCNA蛋白的表达水平逐渐下降。[结论]不同浓度的白藜芦醇对软骨细胞的增殖和Ⅱ型胶原表达有明显的抑制作用。[Objective] We investigated the effect of different concentrations of resveratrol on the proliferation and expression of type α collagen of rat chondrocytes.[Methods] Rat chondrocytes were cultured with different concentrations of resveratrol for 24h.Morphological changes as well as cell proliferation and protein expression levels of both proliferating cell nuclear antigen(PCNA) and Col2a1 were investigated and compared among 5 groups.[Results] Resveratrol clearly inhibited chondrocyte proliferation in new born(within 24 h) Sprague- Dawleyrats,the rate of inhibition was associated with the concentration of resveratrol.Compared with the levels in the control group,expression levels of both PCNA and Col2a1 were downregulated with increasing concentrations of resveratrol.[Conclusion] Different concentrations of resveratrol clearly inhibited chondrocyte proliferationas well asPCNA and type II collagen expression.
