ACE基因I/D多态性对高糖低脂膳食诱导的健康青年血脂比值及血压变化的影响

摘  要:目的探讨血管紧张素转换酶(ACE)基因16内含子插入/缺失(I/D)多态性对高糖低脂(HC/LF)膳食诱导的健康汉族青少年血脂比值及血压变化的影响。方法 56名健康青年志愿者,给予7d平衡膳食和6d HC/LF膳食,分别于第1d、第8d和第14d清晨抽取空腹静脉血,测定血压和血脂水平并计算血脂比值。聚合酶链式反应检测ACE基因16内含子I/D多态性。结果在整体受试人群及性别分层各组中,基础值、HC/LF膳食前、HC/LF膳食后的血脂比值及血压水平在Ⅱ基因型与D等位基因携带者(ID+DD基因型)之间均无统计学差异。与HC/LF膳食前相比,在整体受试人群中,HC/LF膳食后低密度脂蛋白胆固醇/高密度脂蛋白胆固醇(LDLC/HDL-C)和总胆固醇(TC)/HDL-C在两组基因型受试者中均降低;收缩压(SBP)仅在Ⅱ基因型受试者中降低。性别分层分析发现,HC/LF膳食后甘油三酯(TG)/HDL-C和log(TG/HDL-C)仅在女性Ⅱ基因型受试者中升高,TG/HDL-C和log(TG/HDL-C)仅在男性Ⅱ基因型受试者中降低;TC/HDL-C和LDL-C/HDL-C在所有男性受试者和女性D携带者中降低;SBP仅在男性D携带者中降低。结论 HC/LF与ACE基因16内含子I/D多态性I等位基因相互作用可降低男性TG/HDL-C和log(TG/HDL-C)、升高女性TG/HDL-C和log(TG/HDL-C);与D等位基因相互作用可降低男性收缩压、女性TC/HDL-C和LDL-C/HDL-C。Objective To investigate the effects of an insertion/deletion(I/D)polymorphism at the intron 16 of the gene of angiotensin converting enzyme(ACE)on the changes of serum lipid ratios and blood pressure induced by a high-carbohydrate and low-fat(HC/LF)diet in healthy Chinese Han youth.Methods Fifty six healthy Chinese Han young volunteers were enrolled.A washout diet was given for seven days followed by the HC/LF diet for six days.Serum lipids and blood pressure were measured on the 1st,8th,and 14th days.Serum lipid ratios were calculated.The ACEI/D polymorphism was detected by PCR.Results There were no significant differences of serum lipid ratios and blood pressure at baseline and before and after the HC/LF diet between theⅡ genotype and the D carriers(ID and DD genotypes)in the whole study population,the males or the females separately.When compared with those before the HC/LF diet,all the subjects regardless of the genotype experienced statistical decreases of low density lipoprotein-cholesterol/high density lipoprotein-cholesterol(LDL-C/HDL-C)and total cholesterol(TC)/HDL-C,but significant decrease of systolic blood pressure(SBP)was only found in the subjects with theⅡ genotype.After taking into account gender,triglyceride(TG)/HDL-C and Log(TG/HDL-C)decreased in the males with theⅡ genotype and increased in the female counterparts.The decreases of TC/HDL-C and LDLC/HDL-C were observed in all the males and the female D carriers.SBP decreased only in the male D carriers.Conclusion The interaction of the HC/LF diet with the I allele of the intron 16I/D polymorphism at the ACEgene decreases TG/HDL-C and log(TG/HDL-C)in males,but increases TG/HDL-C and log(TG/HDL-C)in females in the Chinese young population.The interplay with the D allele lowers SBP in males,and TC/HDL-C and LDL-C/HDL-C in females.
