摘  要:目的探讨稀释聚维酮碘对体外培养肺癌细胞A549生长的影响,为稀释聚维酮碘在肺癌围术期中的应用提供实验依据。方法选用A549肺癌细胞为研究对象,分别用0.5%、0.25%、0.05%三种稀释度的聚维酮碘在37℃、43℃、47℃下处理对数生长期的肺癌细胞,采用MTT比色法观察不同稀释度的聚维酮碘对肺癌细胞增殖作用的影响。结果三种稀释度聚维酮碘对肺癌细胞A549均具有明显的抑制作用(≥83.62%),43℃、0.05%为聚维酮碘最佳作用条件。在37℃及43℃时,聚维酮碘抑瘤作用明显优于生理盐水,镜下观察可见肺癌细胞发生皱缩、胞核浓缩、靠边等形态学改变。结论研究证实不同稀释聚维酮碘对肺癌细胞A549均有明显抑制作用,43℃、0.05%为其最佳作用条件,从而为聚维酮碘稀释液预防肺癌肿瘤细胞的种植转移提供了一定的实验依据和可行的护理方案。Objective To investigate the effect of diluted povidone-iodine on proliferation and apoptosis of lung cancer A549 and to provide the theoretical basis for its clinical application.Methods Human lung cancer cell line A549 were treated with different dilutions(0.5%,0.25%,0.05%) of povidone-iodine at different temperature(37℃,43℃,47℃) respectively,MTT assay was used to examine the effect of different concentration of diluted povidone-iodine on proliferation of lung cancer cell line A549.Results The proliferation of lung cancer cell line A549 cells in vitro was inhibited by povidone-iodine at the three dilutions(≥83.62%).The optimal combination was povidone-iodine(43℃,0.05%),which could reach the best efficiency.The inhibitive effect of diluted povidone-iodine was significantly better than that of normal saline group at different temperatures(37℃,43℃).Typical apoptotic morphologic changes(shrinkage,concentrated nucleus and side step) of the cell nuclei were observed under the microscopy.Conclusion Diluted povidone-iodine(especially 43℃,0.05%) can strongly inhibit the proliferation of lung cancer cell line A549 in vitro,which would provide a theoretical basis and effective clinical nursing strategies for preventing the metastasis of lung cancer cells.
