摘 要:目的:观察索骨丹水提取物和岩白菜素对3种致炎细胞因子的影响,进而探讨其治疗炎症性疾病的部分机制。方法将体外培养的人单核细胞分为脂多糖(LPS)组、不同浓度的含药组及阴性对照组。除对照组外,各组均加入相同量的脂多糖后继续培养12h,收集上清液,采用ELISA法通过酶标仪测得各组溶液的吸光度值。再计算出各组细胞因子的浓度。结果索骨丹水提物各浓度对IL-1β、IL-6和TNF-α均有显著性抑制作用(P<0.05),其中0.75和1.25mg·mL-1组对IL-6具有极显著的抑制作用(P<0.01)。岩白菜素各组对IL-1β均有显著性抑制作用(P<0.05)。结论索骨丹水提取物可通过减少TNF-α、IL-1β和IL-6的产生和释放而发挥治疗作用,岩白菜素通过抑制IL-1β而发挥抗炎作用。The influence of aqueous extract from Rodgersia aesculifolia Batal and bergenin on cytokines were studied to explore the mechanism of anti-inflammatory.Human monocytes in vitro culture were divided into lipopolysaccharide(LPS) group,the different concentration of medicated groups and the negative control group.Each group were added the same amount of LPS.Collecting supermatant after 12 hours and measuring the absorbance values of each solution by ELISA method.Then computed cytokine concentration of each groups.Every concentration of aqueous extract from Rodgersia aesculifolia Batal could significantly inhibit IL-1β,IL-6 and TNF-α(P<0.05).Among them,0.75mg·mL-1and 1.25mg·mL-1groups could reduce the content on IL-6 very significantly(P<0.01).Every concentration of bergenin could reduce the content on IL-1β(P<0.05).Water extract of Rodgersia aesculifolia Batal can reduce the producing and releasing of TNF-α、IL-1βand IL-6 then play a role of treatment.Bergenin could anti-inflammatory by inhibition on IL-1β.
