大鼠自体原位肝移植后肺组织MD-2的表达及意义

摘  要:目的观察自体肝移植后肺组织病理变化及MD-2、TLR2/4基因和蛋白表达变化。方法选取健康SPF级SD大鼠30只,随机分为假手术对照组(S组,n=6)和移植肝脏再灌注后4、8、16、24小时组(M4、M8、M16和M24组,每组n=6)。观察肺组织病理损伤,Q-PCR法检测MD-2、TLR2/4基因表达,Western Blot法分析MD-2、TLR2/4蛋白表达,ELISA法检测肺组织TNF-α和IL-6浓度。结果假手术组肺组织结构基本正常,移植组表现为肺间质明显出血、充血,中性粒细胞浸润显著增多,以肝脏再灌注后8小时最为明显。与S组相比,M4、M8、M16组肺组织MD-2、TLR2和TLR4基因与蛋白表达显著上调(P<0.05),在肝脏再灌注8小时时达到高峰。与S组比较,M8、M16和M24组肺组织匀浆的TNF-α和IL-6浓度显著升高,在肝脏再灌注8小时时达到高峰。结论大鼠自体原位肝移植术后肺组织MD-2、TLR2和TLR4基因与蛋白表达显著上调,TNF-α和IL-6浓度升高,可能对肝移植急性肺损伤起重要作用。Objective To investigate the gene transcription and expression level of myeloid differentiation protein-2(MD-2) in lung tissues in a rat orthotopic autologus liver transplantation model.Methods Thirty healthy SPF Sprague-Dawley rats were randomly divided into five groups.The rats were divided into sham group(S,n =6),and M4,M8,M16,M24 groups respectively according to the different hours after liver reperfusion(n =6,respectively).Lung pathological morphology changes after liver reperfusion were examined.Q-PCR and western blotting methods were used to detect MD-2 and TLR2 / 4 genes transcription,and protein expression levels.The contents of TNF-α and IL-6 were measured using ELISA kit.Results Normal structure of the lung tissues and seldom inflammatory cells infiltration were found in lung tissues in the sham group,while collapse of the pulmonary structure with severe infiltration of inflammatory cells into the intra-alveolar and interstitial spaces and interstitial edema,most markedly 8h after liver reperfusion in model groups.Compared with the sham group,MD-2and TLR2 / 4 gene and protein expression levels,the concentrations of TNF-α and IL-6 in lung tissues in M4、M8、M16 and M24 groups significantly increased and reached the peak at 8h after liver reperfusion.Conclusion MD-2 and TLR2 / 4 gene and protein expression levels and the concentrations of TNF- αand IL-6 in lung tissues in rat orthotopic autologus liver transplantation models are significantly upregulated,which may contribute to acute lung injury after liver transplantation.
