摘 要:目的观察麻醉药物戊巴比妥钠、水合氯醛在重复麻醉条件下对裸鼠的麻醉效果,研究药物的最佳麻醉剂量以适于磁共振(MR)扫描。方法随机将11只裸鼠按照注射药物和剂量的不同分作5组(H1-H5组)。H1:水合氯醛0.0375ml/10g组;H2:戊巴比妥37.5mg/kg组;H3:戊巴比妥50mg/kg组;H4:戊巴比妥62.5mg/kg组;H5:混合组(水合氯醛0.04ml/10g、戊巴比妥钠50mg/kg)。除混合组每组3只外,其余每组2只。每只小鼠不同时间点重复腹腔麻醉,观察记录麻醉后各项麻醉效果指标。结果水合氯醛和不同剂量的戊巴比妥钠有着不同的麻醉效果,水合氯醛0.0375ml/10g组在注射麻药后的0~18h,小鼠深麻平均时间大概在27.5~60.0min,24h后深麻时间明显延长,接近120min,并且容易死亡;戊巴比妥组重复时间注射麻醉效果均比较稳定,50mg/kg组和62.5mg/kg组深麻期在6~42min间,均未见死亡个例。结论 50mg/kg和62.5mg/kg的戊巴比妥钠麻醉效果相对稳定,后者深麻时间更长,这两个剂量组均可用于磁共振扫描的重复麻醉;而水合氯醛不适宜小鼠的重复麻醉。Objective To observe anesthetic effects of pentobarbital sodium and chloral hydrate which were used repeatly to anaesthetize the nude mice, so that to test the best used anesthesia doses of the two drugs to facilitate the MRI experiment study. Methods 11 nude mice were randomly assigned to 5 groups (H1-H5) according to different injections and doses. H1: 0.0375 ml/10g chloral hydrate group; H2: 37.5?mg/kg pentobarbital sodium group; H3: 50 mg/kg pentobarbital sodium group; H4: 62.5mg/kg pentobarbital sodium group; H5: mixed group (0.04 ml/10g of chloral hydrate, 50 mg/kg of pentobarbital sodium). There were two mice in each group except for the mixed group with three mice in it. Every mouse received intraperitoneal anesthesia at different time points. Evaluation indexes of anesthesia were recorded at the same time. Results There were different anaesthetic effects between pentobarbital sodium and chloral hydrate. The average anesthesia time in depth was about 27.5 - 60 min during 0 -18?h after injection. It was close to 120?min which was significantly prolonged after 24?h. We also found the mice were more likely to die during this time. The anaesthetic effects of pentobarbital sodium groups stayed stable, which anesthesia time in depth was from 6?min to 42 min in 50 mg/kg group and 62.5 mg/kg group, and no death. Conclusion Anaesthetic effects of 50 mg/kg and 62.5 mg/kg for pentobarbital sodium were relatively stable, anaesthesia depth of the latter was much better, which showed that these two kinds of doses could both be used for having an anaesthesia repeatly on MRI, but chloral hydrate was not suitable for it.
