摘  要:目的:观察捏脊对脾虚大鼠海马一氧化氮合酶(nitric oxide synthase,NOS)活力和神经元型一氧化氮合酶(neuronal nitric oxide synthase,nNOS)阳性神经元的影响,探讨其可能机制。方法:通过建立利血平致脾虚动物模型,设立正常组、模型组、药物组、捏脊组。测定大鼠体质量、学习记忆和游泳时间等指标,评定脾虚反应的强度。通过NADPH-d组织化学和nNOS免疫组织化学方法,研究各组大鼠海马NOS活力和nNOS蛋白表达的变化。结果:与药物组和捏脊组相比,模型组大鼠学习记忆能力明显降低,海马结构NADPH-d和nNOS阳性神经元数明显减少(P<0.05)。捏脊疗法使脾虚大鼠的体质量明显增加的同时,使脾虚大鼠的游泳时间明显延长(均P<0.05)。结论:捏脊疗法能有效抗疲劳和改善脾虚大鼠学习记忆能力,其机制可能与捏脊调节脑海马NOS活性和nNOS蛋白表达水平有关。Objective To investigate the effects of chiropractic therapy on actiuity of the nitric oxide synthase(NOS) and the expression level of neuronal nitric oxide synthase(nNOS) in hippocampus of rats with spleen deficiency and explore the underlying mechanism.Method To set up the animal model of splenic asthenia with reserpine.Experimental rats were randomly divided into four groups:the control group,the model group,the Chinese herb group,and chiropractic therapy group.Changes of weight,the ability of learning and memory and swimming time of rats were measured.NADPH-d histochemistry and nNOS immunohistochemistry were used to investigate the changes of NOS and nNOS positive neurons in different groups.Results Chiropractic therapy could increase weight and prolong the swimming time of splenic asthenia rats.Compared with the Chinese herb group and chiropractic therapy group,the ability of learning and memory significantly reduced in the model group.After chiropractic treatment,the ability of learning was improved;number of NOS positive neurons and nNOS positive neurons in hippocampus of splenic asthenia rats were significantly increased compared with the control group(P<0.05 or P<0.01).Conclusion Chiropractic therapy can effectively counteract fatigue and improve the ability of learning memory of rats with splenic asthenia by regulating the content of NOS and nNOS in the hippocampus.
