摘 要:就直接撕取表皮法、牛皮胶印迹法、指甲油印迹法和扫描电镜法等4种常用的气孔观察方法对月季‘卡罗拉’(Rosa hybrida‘Carola’)切花叶片气孔的观察效果进行了比较.结果表明,扫描电镜法最适合该切花叶片气孔的观察,而其他3种观察方法均存在一些不足和局限性.进一步采用扫描电镜法对该切花不同部位气孔分布的观察发现,该切花的气孔主要分布在叶片下表皮,萼片下表皮也有一定数量的气孔分布,叶柄和花茎的表皮则仅有零星气孔分布,而叶片上表皮、萼片上表皮、花托、花瓣上表皮和花瓣下表皮均未见气孔分布.另外,通过对该切花叶片下表皮和萼片下表皮的气孔特征进行的分析表明,前者的气孔密度为后者的3倍,且前者的气孔面积也显著大于后者.Four common methods for plant stomatal observation , including directly shredding , oxhide ge-latin blotting, nail polish blotting and scanning electron microscopy (SEM), were used to observe stoma-ta in leaves of cut roses ( Rosa hybrida‘Carola’ ) , and the results showed that SEM was the most appli-cable to observing stomata of this cut flower , while the other 3 methods were not applicable due to their shortcomings and limitations .Observation on stomatal distribution in different parts of this cut flower by SEM indicated that its stomata mainly distribute in lower epidermis of leaf , and with small amount of sto-mata in lower epidermis of sepal and with sporadic stomata in leafstalk and stem surface , however with no stomata in upper epidermis of leaf , upper epidermis of sepal , receptacle , upper epidermis of petal and lower epidermis of petal .Further investigation of the stomatal characteristics in lower epidermis of leaf and lower epidermis of sepal showed that the stomatal density of the former is 3 times higher than that of the latter , and that the stomatal area of the former was also significantly larger than that of the latter .
