骨髓间充质干细胞自体移植及丹参注射液对大鼠冷冻伤脑水肿模型的长期疗效观察

摘　　要：目的观察骨髓间充质干细胞自体移植及丹参注射液对大鼠冷冻伤脑水肿模型神经功能改善的长期影响。方法将Wistar大鼠自体骨髓MSCs在体外扩增并经Brdu标记后,通过颈内动脉注射将其植入Wsitar大鼠冷冻伤脑水肿模型体内,同时使用丹参注射液腹腔注射进行治疗,进行为期一年的疗效观察。结果经颈内动脉注射的大鼠MSCs细胞,结合丹参注射液的治疗作用,B组Brdu阳性细胞的数量和神经功能恢复的程度高于A组,且有明显统计学差异(P<0.05)。但阳性细胞的数量随时间推移较移植初期有明显下降,大鼠神经功能评分随时间推移无明显变化。结论骨髓MSCs自体移植结合丹参注射液治疗对大鼠冷冻伤脑水肿模型可发挥长期治疗作用,疗效稳定。Objective To explore the long curative effect of autologous transplantation of MSCs and traditional Chinese medicine like Dansen on severe brain injury induced by cold .Methods After MSCs from the Wistars were proliferated and labelled with Brdu in vitro ,the autologous MSCs were transplanted into the cold-induced brain edema model through the internal carotid artery with salvia intraperitoneal injection .Results The MSCs from the Wistars can immigrate into the brain injury area through the internal carotid artery .The number of Brdu-positive cells in the in-jured brain areas and the neurological scores in theMSCs transplatation groups (group B) were significantly more than that in the control group(group A) (P<0.01) .As times went by the neurological scores in the group B have no significant changes .Conclusion The MSCs labelled with Brdu exert the curative effect with salvia intraperitoneal injection ,which no significant decrease as times go by .
