摘  要:目的探讨不同浓度气道给氧对急性百草枯大鼠模型肺组织氧化应激相关指标的影响。方法 160只SD大鼠随机分为正常对照组(A组)、模型组(B组)、30%浓度的给氧组(C组)和50%浓度的给氧组(D组),每组40只。B、C、D组以50 mg/kg百草枯(稀释于1 mL生理盐水)一次性灌胃染毒造模1 h后,C、D给氧组通过气道给予相应浓度氧治疗,6 h/d。A组和B组正常吸入空气。分别于第1、3、7、14、21天每组各取8只SD大鼠,麻醉后腹主动脉采血1 mL立即行血气分析及血LAC检查,随即放血处死。留取右肺测湿/干重比,选取3 d右肺上叶组织用10%甲醛固定,用于病理组织学检查;左肺制成匀浆,用于检测肺组织匀浆的过氧化氢酶(CAT)活力和SOD水平。结果 HE染色结果显示C组淤血、水肿现象减弱程度最为明显。与B组各时间点比较,C、D组组织匀浆中SOD和CAT的水平均有不同程度的升高,动脉血中PO2上调,PCO2、LACP下调,差异均有统计学意义(P<0.05或P<0.01);但C、D组比较差异无统计学意义。与B组各个时间点比较,C、D组肺部组织湿/干(W/D)比值均有不同程度的下调,差异有统计学意义(P<0.05或P<0.01)。结论气道给氧可减弱百草枯致大鼠急性肺损伤的氧化应激反应,对肺组织有一定的保护作用,以30%氧浓度效果较好。Objective To investigate the effects of different concentrations of oxygen through airway on oxidative stress-related indicators of lung tissue in paraquat intoxicated rats. Methods 160 SD rats were randomly divided into normal control group( group A),model group( group B),30%oxygen concentration( group C) and 50% concentration oxygen group( group D),each had 40 rats. B,C,D groups were given 50 mg /kg paraquat( diluted in 1 mL saline) disposable by intragastric administration. After 1h,oxygen therapy through airway were given to group C and D,6h /d; group A and normal group B intake air. 8 rats were anesthetized and 1 mL blood were drawn through abdominal aorta at 1,3,7,14,21 d,and blood gas and blood LAC were immediately analyzed and checked,after that all rats were bled to death. The wet /dry weight ratio of right lungs were measured and lobe tissue in 3d were fixed with 10% formalin for histological examination; left lungs were homogenizied and catalase( CAT) activity and SOD levels in lung homogenates were detected. Results HE staining results showed that the congestion,edema were weakened obviously in group C. Compared with group B at each time point,SOD and CAT levels in tissues were up-regulate with varying degrees in C,D groups,arterial PO2 raise,PCO2,LACP downregulate,and the differences were statistically significant( P < 0. 05 or P < 0. 01),but there were no significant differences between C,D groups. Compared with group B at each time point,the wet /dry( W /D) ratio of lung tissues were decreased at different degrees in C,D groups,and the difference was significantly( P < 0. 05 or P < 0. 01). Conclusion Giving a certain concentration of oxygen through airway can reduce oxidative stress in lung tissue,protect lung tissue in paraquat intoxicated rats,and effect of 30% oxygen concentration is remarkable.
