摘  要:目的探讨丹参川芎嗪注射液对妊娠期高血压大鼠模型的效果及对胎盘细胞CyclinB1、p21蛋白表达量的影响。方法将64只孕鼠随机分为4组:正常妊娠大鼠作空白对照,妊娠高血压大鼠作阴性对照,硫酸镁治疗组作阳性对照,丹参川芎嗪注射液组为实验组;每组16只。妊娠14 d时,尾静脉注射亚硝基左旋精氛酸甲酯建立妊娠期高血压模型。从妊娠第14天开始给药,空白对照组及阴性对照组均给予与药物同剂量的生理盐水;阳性对照组及实验组给予硫酸镁及丹参川芎嗪注射液治疗,3 d为一个给药疗程。通过大鼠套尾法测量血压,尾静脉取血法测定24 h尿蛋白,并观察各组妊娠大鼠所生新生鼠的身长、体重、死胎率、后肢畸形率和胎盘重量,并通过免疫蛋白印迹法测定各组大鼠胎盘细胞中CyclinB1、p21蛋白表达量。结果与空白组相比,阴性对照组孕鼠尿蛋白及血压明显增加,新生鼠的身长、体重和胎盘重量均显著降低,死胎率以及畸形率显著增高(P<0.05);与阴性对照组相比,阳性对照组和实验组孕鼠收缩压及尿蛋白浓度均显著降低,新生鼠的身长、体重和胎盘重量均显著升高,死胎率以及畸形率显著减少(P<0.05);阳性对照组及实验组各指标差异无统计学意义。与空白对照组比较,阴性对照组孕鼠CyclinB1蛋白浓度显著下降(P<0.05),p21浓度显著增高(P<0.05);与阴性对照组相比,阳性对照组和实验组孕鼠CyclinB1蛋白浓度显著升高(P<0.05),p21浓度显著降低(P<0.05);阳性对照组与实验组相比较,差异无统计学意义。结论川芎嗪注射液能够显著降低妊娠高血压孕鼠的死胎率以及畸形率,改善新生鼠状况,并且能够上调cyclinB1以及下调p21表达,这可能是川芎嗪注射液治疗效果的原因之一。Objective To investigate the effects of salvia injection on the effect of TMP rat model of hypertension in pregnancy and placental cells CyclinB1,p21 protein expression. Methods 64 rats during pregnancy were divided into four groups: normal pregnant rats as control,pregnancy-induced hypertension rats as negative control,pregnancy-induced hypertension rats treated with magnesium sulfate as positive control,pregnancy-induced hypertension rats treated with salvia ligustrazine injection as experimental group,each had 12 rats. Control group and negative group rats were given the same dose of the drug saline; positive group and the experimental group were separately given magnesium sulfate and salvia ligustrazine injection therapy,three days a dosing regimen. Blood pressure in rats tails were measured,24 h urine protein though tail vein blood were determined,newborn rats weight,still birth rate,limb deformity rate and placental weight of pregnant rats in each group were observed,and placental cell CyclinB1,p21 protein expression were detected by Western blott. Results Compared with control group,blood pressure and urine protein of pregnant rats in negative control group,were increased significantly( P < 0. 05),neonatal height,weight and placental weight were decreased significantly( P < 0. 05),and the stillbirth rate and malformation rate was significantly higher( P < 0. 05). Compared with negative control group,blood pressure and urine protein of pregnant rats in positive control group and experimental group were decreased significantly( P < 0. 05),neonatal height,weight and placental weight were increased significantly( P < 0. 05) and the stillbirth rate and malformation rate was significantly lower( P < 0. 05). There were no significant differences of those indexes between positive control group and experimental group. Compared with control group,cyclinB1 protein expression in negative control group was decreased( P < 0. 05),and p21 was significantly increased( P < 0. 05),Compared with negative control group,cyclinB
