摘 要:本试验采用PCR产物直接测序法寻找家兔细胞分裂周期蛋白25同源蛋白C(Cdc25C)基因SNPs位点,并用PCR-RFLP分型技术对新西兰兔共174个个体进行遗传多态性分析。结果表明,在家兔Cdc25C基因编码区1296bp处发生同义突变(c.1296 T>C),该突变位于Cdc25C蛋白的RHOD(rhodanese)结构域,TT基因型为优势等位基因型(0.6437),T等位基因为优势等位基因(0.7874)。遗传分析表明,c.1296 T>C位点在新西兰兔中的杂合度(He)为0.3348,有效等位基因数(Ne)为1.5033,多态信息含量(PIC)为0.2788,表现为中度多态(0.25<PIC<0.50),表明该位点有较高的遗传变异,选择潜力较大。The single nucleotide polymorphisms(SNPs) of Cdc25C gene were firstly detected by directing sequencing, and then the candidate SNP of Cdc25C gene was genotyped by PCR-RFLP in a total of 174New Zealand rabbits. The result showed that one SNP(c.1296 T>C) located in coding region of Cdc25C gene was detected, which was also located in the rhodanese(RHOD) domain; TT genotype of c.1296 T>C was the dominant genotype(0.6437) and T allele was the dominant allele(0.7874). The genetic analysis of c.1296 T>C in the New Zealand rabbit showed that heterozygosity(He) was 0.3348, effective number of alleles(Ne) was 1.5033, and polymorphic information content(PIC) was 0.2788 which showed genetic diversity was moderate polymorphism(0.25<PIC<0.50). The genetic diversity indicated relatively high genetic variation and a selective potentiality in NZW, which could be applied with marker-assistant selection to improve the molecular breeding on breeding traits.
