摘 要:为了分析免疫仔猪外周血T淋巴细胞CD28表达特点,采集接种蓝耳病弱毒疫苗和猪瘟弱毒疫苗的仔猪前后3d外周血液,并分离外周血单核细胞,采用实时定量PCR法检测仔猪外周血中T细胞CD28基因的相对表达量。结果显示仔猪在接种蓝耳病、猪瘟疫苗前后,CD28的表达有明显的提高,猪免疫前后CD28表达变化与机体接受抗原刺激有关。接种疫苗后,CD28在细胞内的表达明显增高,但与免疫的疫苗种类没有直接关系。To study the expression characteristics of CD28 on T-lymphocyte in piglet.Peripheral blood were collected from piglet vaccinated with attenuated PRRS virus vaccine,classical swine fever virus vaccine at pre and post three days.The segregated peripheral blood monouclear cell were reversed transcription using primer of Oligo(dT),and the relative expression of CD28 in T cells of peripheral blood were detected by Real-time quantitative PCR.The expression of CD28 increased greatly after immune the pre-immune attenuated PRRS virus vaccine and classical swine fever virus vaccine.The expression of CD28 gene in piglet changed after immune associated with the body antigen stimulation.After vaccination,the expression of CD28 on cells increased obviously,but had no direct relevance with the types of vaccine immune.
