摘  要:首先采用蛋白印迹以及激光共聚焦显微观察的方法,研究猪瘟病毒感染对细胞内自噬相关蛋白LC3表达及定位的影响;然后用自噬诱导剂雷帕霉素或自噬抑制剂3-甲基腺嘌呤处理猪肾细胞,通过病毒滴度测定研究细胞自噬对病毒复制的影响。发现在病毒感染24h后自噬相关蛋白LC3由LC3-Ⅰ酯化为LC3-Ⅱ,并观察到病毒感染的细胞中出现了LC3蛋白聚点;细胞自噬体的诱导能显著增加病毒滴度。本研究表明猪瘟病毒在感染细胞后能诱导细胞自噬,该细胞自噬过程有利于病毒的增殖。To demonstrate the relationship between autophagy and CSFV,PK-15 cells were infected with classical swine fever virus(CSFV),and the expression and aggregation of autophagy-related protein LC were analyzed by western-blot and confocal microscopy.Furthermore,PK-15 cells were treated with autophagy inducer rapamycin or autophagy inhibitor 3-methyladenine,and the effects on CSFV replication were evaluated by virus titration.The results showed that autophagy related proteins LC3-I was converted to LC3-II at 24hpost-infection and LC3 punctuates were observed by confocal microscopy in virus infected cells.After treatments with rapamycin,it was found that the induction of autophagy significantly increased the virus titers.Taken together,the data here indicate that autophagy can be induced by CSFV infection and benefit the virus replication in turn.
