摘 要:分别以活菌浓度为109(A组)、106(B组)ccu/mL的(MG)F株及生理盐水(C组)点眼接种SPF鸡,采用流式细胞技术及ELISA方法对免疫前后外周血淋巴细胞(PBL)中Bu-1a+B、TCRγδ+CD3+T细胞比例及血清IgM、IgG、IgA的动态变化规律进行研究。结果显示各组Bu-1a+B/PBL于免疫后5、7d由高至低依次为A、B和C组,且各组差异显著(P<0.05);A、B组TCRγδ+CD3+T/PBL于免疫后3d略有升高且比值较接近,都高于C组,但差异不显著(P>0.05);各组IgM于免疫后7、14和21d差异显著(P<0.05),IgG于免疫后14、21和28d差异显著(P<0.05),IgA于免疫后28d差异显著(P<0.05),由高至低依次为A、B、C组。研究结果表明,免疫(MG)F株可较好的提高Bu-1a+B、血清IgM、IgG、IgA介导的体液免疫作用,且(MG)F株活菌浓度与Bu-1a+B/PBL比值、血清IgM、IgG、IgA浓度呈正相关;但免疫(MG)F株不能显著提高鸡外周血TCRγδ+CD3+T/PBL比值,且(MG)F株活菌浓度与TCRγδ+CD3+T/PBL比值无明显相关性。To evaluate the effects of mycoplasma gallisepticum(MG)F vaccine infection on Bu-1a+B,TCRγδ+CD3+T in peripheral blood lymphocyte(PBL)and IgM,IgG,IgA in serum of specefic pathogen free(SPF)chickens,SPF chickens were inoculated with 109(A group),106 ccu/mL(MG)F vaccine(B group)and saline(C group),respectively.In addition,the change of Bu-1a+B/PBL,TCRγδ+CD3+T/PBL,IgM,IgG and IgA were analyzed by flow cytometrer and ELISA.The results showed that Bu-1a+B/PBL inorder from high to low was A,B and C group with significant differences on 5and 7day post vaccination(dpv)(P<0.05).TCRγδ+CD3+T/PBL of A and B group increased slightly and was higher than C group with no significant differences(P>0.05)after 3dpv.IgM on 7,14,21 dpv,IgG on 14,21,28 dpv and IgA on 28 dpv inorder from high to low was A,B and C with significant differences(P<0.05).These findings indicated that(MG)F vaccine could reinforce humoral immunity by Bu-1a+B/PBL,IgM,IgG and IgA which was direct proportion with the inoculation dose of(MG)F vaccine.But(MG)F vaccine could not reinforce TCRγδ+CD3+T/PBL which was no correlation with the inoculation dose.
