摘  要:为研究不同浓度的金黄色葡萄球菌对奶牛乳腺成纤维细胞(BMFB)血小板源性细胞生长因子(PDGF-BB)及其受体PDGFRβ表达的影响,采用不同浓度的热灭活金黄色葡萄球菌(0、105、106和108 CFU/mL)作用于BMFB细胞,分别于不同的时间点(6、12、24和48h)采用RT-PCR法检测PDGF-BB及PDGFRβmRNA表达,Western-blot法检测PDGF-BB及PDGFRβ蛋白的表达。结果显示,金黄色葡萄球菌作用于BMFB后,PDGF-BB mRNA表达量的升高呈明显的时效关系和量效关系(P<0.05);PDGF-BB蛋白表达升高呈明显的时效关系(P<0.05)。PDGFRβmRNA的表达量较对照组升高,呈正量效关系和正时效关系,在12h达到峰值后下降,同时mRNA表达量呈负量效关系(P<0.05);PDGFRβ蛋白表达量呈正时效关系(P<0.05)。本研究证实了金黄色葡萄球菌可以上调BMFB PDGF-BB及PDGFRβmRNA和蛋白的表达。In order to explore the effect of heat-inactivated Staphylococcus aureus(S.aureus)on the mRNA expression and protein secretion of PDGF-BB and PDGFRβin bovine mammary fibroblasts,the bovine mammary fibroblasts cultured in vitro were exposed to the heat-inactivated S.aureus at 105,106,108 CFU/mL,respectively,and the total RNA and protein were extracted from the cells in different time(6,12,24,48h)for examination by real-time RT-PCR and Western-blot.The results showed that the mRNA expression of PDGF-BB in treated cells is in a time and dosedependent manner and the protein levels of PDGF-BB in treated cells rises in a time-dependent manner(P<0.05).mRNA expression of PDGFRβrises in a time and dose-dependent manner after stimulated by S.aureus(P<0.05).Meanwhile,the mRNA expressions of PDGFRβis significantly elevated when BMFB were exposed to S.aureus,and reaches the maximal levels at 12 h,respectively,and then expression goes down.The protein secretion of PDGFRβis in a time-dependent manner(P<0.05).In conclusion,the results indicated that heat-inactivated S.aureus up-regulated the expression of PDGF-BB and PDGFRβmRNA and protein in bovine mammary fibroblasts.
