猪血凝性脑脊髓病毒刺激小鼠树突状细胞成熟及炎性因子分泌

摘　　要：为了了解猪血凝性脑脊髓炎病毒(Procine hemagglutinating encephalomyelitis virus,PHEV)感染机体后对树突状细胞(Dendritic cell,DC)的激活状况,以及DC在PHEV感染过程中参与免疫应答的能力,本试验分离了BALB\c小鼠骨髓细胞,并通过条件培养基诱导培养8d,获得纯度约77.69%的DC。将PHEV接种DC后,分别对其表面成熟标志分子CD40、CD80、MHCⅡ类分子以及分泌炎症因子的能力进行检测分析。结果显示,PHEV感染DC后T细胞共刺激分子CD40、CD80及MHCⅡ均有一定程度的上调,同时DC分泌细胞因子和趋化因子的能力显著增强,尤其IL-1β的mRNA表达量增高近10倍。经荧光定量PCR方法检测病毒刺激后DC模式识别受体的表达情况结果显示,TLR4和TLR9表达量变化不明显,TLR7的表达受到抑制;但Rig-I的表达量显著增加。结果表明,PHEV在体外能有效的活化DC并促使其成熟为有效的抗原提呈细胞,具有潜在的激活T、B淋巴细胞的能力。For the purposes that are whether mouse dendritic cells(DCs)mature post infection of porcine hemagglutinating encephalomyelitis virus(PHEV)and what is the ability of antigen presentation,we acquired DCs of which purity is about 77.69% by separation and culture of bone marrow cells with conditioned medium for 8days.Expression of surface maturation markers such as CD40,CD80 and MHCⅡ,inflammatory cytokinesis and chemokine were detected and analysed after infection of PHEV by qPCR and FACS.The results showed that T cell co-stimulatory molecules CD40,CD80,CD86 and MHCⅡ upregulated,but mRNA expression of inflammatory cytokines and chemokine also increased obviously,especially for IL-1β.The change of TLR4 and TLR9was not significantly post infection,and TLR7 were inhibited while Rig-I has a considerable increased by analyzed qPCR.Thus,we conclude that PHEV can effective activate DCs and prompt its mature in vitro.
