摘 要:为研究鹅不同组织生长激素受体(GHR)和胰岛素样生长因子-I(IGF-I)基因发育性变化,本试验选择0日龄杂交鹅(吉林白鹅×农安籽鹅)78只作为试验动物,应用荧光定量技术分别对0、14、28、42、56日龄鹅心脏、肝脏、腺胃、腿肌GHR以及IGF-I mRNA进行测定。结果显示,0~56日龄期间杂交鹅GHR以及IGF-I mRNA在肝脏中表达量最高,其次为腿肌和腺胃,心脏GHR以及IGF-I mRNA表达量最低。鹅心脏、肝脏、腺胃和腿肌GHR mRNA表达量随日龄增加有极显著的发育性变化(P<0.01)。鹅心脏、肝脏、腿肌GHR mRNA表达量均于14日龄达到峰值,鹅腺胃GHR mRNA表达量于28日龄达到峰值。鹅肝脏、腺胃和腿肌IGF-I mRNA表达随日龄增加有极显著的发育性变化(P<0.01)。鹅心脏、腿肌IGF-I mRNA表达量于28日龄出现峰值,但肝脏、腺胃IGF-I mRNA表达量在0~56日龄不断升高。总体上看,公鹅与母鹅各组织GHR以及IGF-I mRNA表达量无显著差异(P>0.05)。结果表明,日龄对于GHR和IGF-I基因在鹅各组织表达具有重要影响;性别对鹅组织GHR和IGF-I基因表达影响较小;鹅各组织GHR和IGF-I基因发育性变化具有组织特异性,各基因表达量在鹅不同组织出现峰值的时间并不一致。The objective of this study was to investigate the developmental changes of expression of GHR and IGF-I genes in Geese.Seventy-eight cross breed geese(0-day-old)were used in this experiment.The expression of GHR and IGF-I mRNA in heart,liver,glandular stomach and leg muscle of geese were detected on d 0,14,28,42 and 56by qRT-PCR.The results showed that the expression of GHR mRNA was the highest in the liver of geese from d 0to 56,following leg muscle and glandular stomach,and the lowest in heart.The expression of GHR mRNA in heart,liver,glandular stomach and leg muscle of geese were significantly changed with increasing age(P<0.01).The peak of GHR mRNA expression in heart,liver and leg muscle was on d 14,however,that of glandular stomach on d 28.The expression of IGF-I mRNA in liver,glandular stomach and leg muscle of geese were significantly changed with increasing age(P<0.01).The peak of IGF-I mRNA expression in heart and leg muscle was on d 28,however,that of liver and glandular stomach increased with the increasing age from d 0to 56.No significant difference was found between males and females in IGF-I mRNA expressions of heart,liver,glandular stomach and leg muscle(P>0.05).The results indicated that,developmental ages had important influences on the expressions of GHR and IGF-I genes in different tissues of geese,however,sex had less effect.The developmental changes of GHR and IGF-I gene expressions have tissue specificity,the the expressions of peak of two genes in different tissues of geese were not consistent.
