摘 要:为研究乳香提取物对NIH-3T3细胞的增殖、细胞周期和ERK1/2蛋白表达及p-ERK1/2蛋白表达的影响,将NIH-3T3细胞传代后,随机分为药物干预组和空白对照组。在药物干预组向NIH-3T3细胞中加入不同浓度的乳香提取物并培养24h,利用CCK-8法检测细胞的增殖率,利用流式细胞术检测细胞周期比例,利用Western-blot检测ERK1/2蛋白及p-ERK1/2蛋白的表达。结果,分别用4×10-1 g/L和8×10-2 g/L乳香提取物干预细胞24h,吸光度较对照组差异极显著(P<0.01)。用流式细胞术检测细胞周期,乳香提取物质量浓度为4×10-1 g/L、8×10-2 g/L和1.6×10-2 g/L时,S期比例较对照组差异极显著(P<0.01),且随着乳香提取物质量浓度的降低,细胞周期中S期百分比降低,呈现出剂量依赖性。Western-blot结果显示,ERK1/2蛋白与对照组相比没有发生明显变化;但p-ERK1/2与对照组相比,随着乳香提取物的质量浓度升高,p-ERK1/2增高,且呈剂量依赖性。结果表明,乳香提取物通过激活ERK1/2信号通路促进NIH-3T3细胞的增殖。We studied the effects of frankincense extract on NIH-3T3 cell proliferation,the cell cycle and expression of ERK1/2and p-ERK1/2.We took the NIH-3T3 cell in vitro as the object of the research.After passage,NIH-3T3 cells were randomly divided into drug intervention group and control group.In the drug intervention group,NIH-3T3 cell was treated respectively with frankincense extract at different concentration for 24 h.The effects of frankincense extract on cell growth were detected using CCK-8method,the cell cycle of NIH-3T3 cell was detected by flow cytometry and the expression of ERK1/2and p-ERK1/2was detected by Western-blot.In result,when drug concentration was 4×10-1 and 8×10-2 g/L after treating cells for 24 h,there was a extremely significant difference on absorbance compared with the control group(P<0.01).Cell cycle detected by flow cytometry also proved that there was extremely significant difference on percentage of S-phase compared with the control group(P<0.01)when drug concentration was 4×10-1 g/L,8×10-2 g/L and 1.6×10-2 g/L after treating cells for 24 h.After treating cells with different concentrations of frankincense extract for 24 h,we detected the expression of protein by Westernblot.Compared with the control group,the ERK1/2 expression had no significant change,but the expression of p-ERK1/2was increased with the increase of drug concentration in a dose-dependent manner.
