摘  要:为了研究黄芪总黄酮(TFA)的体外抗炎作用,通过脂多糖体外诱导小鼠RAW264.7细胞建立炎症模型,采用MTT法检测黄芪总黄酮的细胞毒性作用,用ELISA检测细胞上清液中IL-6、IFN-γ、IL-1β及TNF-α水平,采用Griess法检测细胞内NO表达水平。MTT结果显示,10~100μg/mL TFA对RAW264.7细胞没有明显的毒性作用。与模型组比较,10、25、100μg/mL TFA均能抑制RAW264.7细胞过量分泌IL-6、IFN-γ、IL-1β、TNF-α及NO,且呈一定的量效关系。本试验结果表明黄芪总黄酮具有体外抗炎活性。To observe the in vitro anti-inflammatory effect of total flavonoids of Astragalus(TFA),lipopolysaccharide was used to induce RAW264.7cells to build model of inflammation.The effect of TFA on cell toxic was detected by MTT.The levels of IL-6,IFN-γ,IL-1βand TNF-αin culture fluid were detected by ELISA,and level of NO was detected by Griess method.MTT results showed that 10 to 100μg/mL TFA had no obvious toxicity on RAW264.7cells.Compared with the model group,the mass concentrations of IL-6,IFN-γ,IL-1βand TNF-αand the molarity of NO were inhibited by 10,25 and 100μg/mL TFA,respectively,in a dose-dependent manner.In conclusion,TFA has in vitro anti-inflammatory activity.
