摘 要:【目的】探讨黄肉猕猴桃发育过程中果实类胡萝卜素合成途径中相关差异基因的功能。【方法】以黄肉型中华猕猴桃‘金丰’果实为试材,在前期RNA-seq测序的基础上,利用实时荧光定量PCR技术检测其类胡萝卜素合成过程中相关差异基因的表达水平。【结果】Unigene23885、CL10798、Unigene11266、CL1511四个基因相对表达量较高,其他基因表达量相对较低;相关性分析结果显示,类胡萝卜素含量与Unigene11266、Unigene23885、Unigene2673三个基因相对表达量显著相关,与基因Unigene23823、Unigene12775相对表达量呈极显著相关。聚类分析结果显示,基因Unigene23885和Unigene11266、基因CL1511和CL10798分别被聚为一类,其表达模式非常接近且与其他基因明显不同,并在类胡萝卜素合成中高水平表达,基因CL10467与其距离最近。【结论】综合相关性分析和聚类分析,基因Unigene11266、Unigene23885可能是类胡萝卜素合成的关键基因,且较其他基因而言,基因CL10467、Unigene12775、CL10798、CL1511、Unigene2673与其关系更密切。【Objective】To explore the role of related difference genes in the carotenoid biosynthesis in yel-low-fleshed kiwifruit.【Methods】Based on RNA-seq,a yellow flesh cultivar‘Jinfeng'(Actinidia chinensis)was used to explore the carotenoid synthesis related gene expression through q PCR technique.【Re-sults】Four genes(Unigene23885,CL10798,Unigene11266 and CL1511)had much stronger expres-sion,and correlation analysis showed a significantly positive correlation existed between the carotenoidcontents and four gene(Unigene11266,Unigene23885,Unigene2673)(P< 0.05) and Unigene23823,Unigene12775(P<0.01) expressions. The cluster analysis showed that Unigene23885 and Unigene11266,gene CL1511 and CL10798 were clustered together and belong to the same category,but differ-ent from other genes as they had the same expression pattern in which it was significantly different fromother genes,gene CL10467 was nearest with them.【Conclusion】Based on all the above results,Unige-ne11266 and Unigene23885 might be the key genes during the carotenoid synthesis in yellow flesh kiwi-fruit,and the genes CL10467,Unigene12775,CL10798,CL1511 and Unigene2673 had a closer rela-tionship carotenoid synthesis,compared with other genes.
