摘  要:以被去除脾的昆明小白鼠为实验动物(40只),接种牛源环形泰勒虫吐鲁番流行株Ta-t,进行小鼠体内扩繁试验,并于接种后第8、12、24、48小时以及第3~10天每天经血液涂片检查接种后昆明小白鼠的红细胞染虫率,观察分析其扩繁情况;经昆明小白鼠心脏、眼球采血的方法收集其最高染虫率(15%以上)的血液内裂殖子,并进行冻存,摸索其保存、复苏条件等。结果显示,除脾术后昆明小白鼠均存活,检测其呼吸、脉搏、食欲、精神状态皆良好,说明摘脾手术成功;接种后第48小时可在昆明小白鼠血液里观察到裂殖子,第5~9天每组昆明小白鼠的平均染虫率可达15%~20%(最高峰),第10天后染虫率开始下降,血液涂片中裂殖子数量渐渐减少;经试验发现,在雌雄性昆明小白鼠体内的染虫率差异极显著(P=0.007<0.01),去脾昆明小白鼠与未去脾昆明小白鼠的染虫率差异也极显著(P=0.000 2<0.01),牛源环形泰勒虫在除脾的昆明小白鼠血液内能被置换、增殖。染虫率为15%以上时昆明小白鼠血液里的裂殖子可用冷冻(-80℃)保存,亦可制作玻片裂殖子抗原备用,保存半年以内的裂殖子复苏活性最佳。该试验数据可为建立环形泰勒虫动物模型、教学、科研提供丰富的血液原虫株资源。The splenectomized Kunming mice(n=40)were used as experimental animals,and inoculated with the Turpan epidemic strain Ta-t of Theileria annulatafrom cattle,and the in vivo propagation experiment was carried out.The infectious rate of erythrocyte and propagation conditions were observed at hour 8,12,24,48 and on day 3to 10post-inoculation by blood smears test.Merozoites of blood in maxium parasitemias(over 15%)were collected through the way of heart and eyeball blood sampling and then cryopreserved,stored and anabiosis.In result,all mice survived after operation and their breath,heart beat,appetite and mental state were well,which proved that the splenectomized operation was successful.Merozoites can be observed in the mice's blood at hour 48post-inoculation and the average infection rate of erythrocyte can reach 15% to 20% between 5dand 9d,and the percentages begin to decrease after 10 dand merozoites can not be observed in blood.The propagation rates of male and female have an significant difference(P=0.007< 0.01),and a difference of infection rate was found between the splenectomized and nonsplenectomized Kunming mice(P =0.000 2<0.01).The bovine T.annulata can be replaced and proliferated in splenectomized Kunming mice.The merozoites in the blood of Kunming mice with the infectious rate over 15% can be cryopreserved and also be made into slide merozoites antigen.The recovery activity of merozoite is best in half a year.This study can lay the foundation for establishing T.annulata models and providing rich strains for teaching and research.
