广东某奶牛场蓝舌病病毒分离株血清型与基因型的鉴定

摘　　要：为了解广东省蓝舌病病毒(BTV)流行的主要血清型及基因型,于2013-2014年在广东省汕头市某奶牛场进行了蓝舌病病毒监测及病毒分离,同时扩增了分离株的VP7和VP2基因,采用生物信息学软件进行序列分析并构建系统进化树。结果显示,从64份BTV抗体阳性牛、山羊血液中分离到16株BTV,主要血清型为BTV-2、BTV-4、BTV-12和BTV-16。遗传进化分析表明,其中3个BTV-2分离株与我国台湾(AY493687)、日本(AB686224)、澳大利亚(JQ240322)的分离毒株的遗传关系较近;8个BTV-4分离株与1997年分离自我国云南的毒株(JX560414)同属一个进化分支;2个BTV-12分离株与印度毒株(KC662613)位于同一分支;4个BTV-16分离株与日本毒株(AB686225和AB686226)同在一个大分支上。本研究结果为丰富我国蓝舌病的流行病学资料和疫苗研制奠定了基础。To characterize bluetongue virus(BTV)isolated from Guangdong Province,virus isolation,amplification and sequence analysis of the specific target gene of VP7 and VP2proteins of BTV were carried out.Results showed that a total of sixteen BTV strains from four different serotypes(BTV-2,4,12and16)were isolated.Genetic relationship of three BTV-2strains were close to the strains from Taiwan(AY493687)of China,Japanese(AB686224)and Australia(JQ240322).Eight BTV-4strains and a strain(JX560414)from Yunnan Province of China belonged to the same branch.Two BTV-12 strains were on the same branch with the India strain(KC662613).There was a close relationship between four BTV-16 strains and Japanese strains(AB686225and AB686226).This study provided useful information for the epidemiology of bluetongue in China,and laid the foundation for bluetongue vaccine development.
