摘  要:目的:研究交感神经在小鼠子宫局部免疫中的作用.方法:昆明种雌性小鼠,实验组连续5d腹膜腔注射6-羟多巴胺(6-OHDA)进行交感神经阻断实验,分别于注射后第1、3、5、7、9、14天取子宫;所有小鼠腹膜腔注射6-OHDA阻断交感神经,之后实验组每天腹膜腔注射5、10、25 μg去甲肾上腺素(NE)进行NE处理实验,在注射的第5天取子宫.免疫组织化学检测子宫巨噬细胞膜受体CD14和CD204的表达.结果:与对照组相比,交感神经阻断后子宫内膜、肌层和外膜CD14+巨噬细胞均显著增多,CD14表达显著上调;对照组和实验组第1天未检测到CD204的表达,其余各组均有CD204的表达,且表达量随着时间推移而增多,第9天达到高峰.各实验组注射NE后,子宫CD14+、CD204+巨噬细胞显著减少,CD14、CD204表达显著下调,中、高浓度组下调较明显.结论:交感神经及其递质NE可以通过调节巨噬细胞数量和膜受体CD14、CD204的表达参与子宫局部免疫状态的维持.Objective:To investigate the roles of sympathetic nerve and its transmitter in mouse local immunological reaction.Methods:Female mice of Kunming were used.6-hydroxydopamine(100 mg/kg) was peritoneally injected continuously for 5days to block sympathetic effect.The mice uteri were extracted separately on day 1,3,5,7,9 and 14.All the mice were injected with 6-hydroxydopamine to block the sympathetic effect,and then for experimental group,two injections were performed with norepinephrine(NE) with the doses of 5,10 and 25 μg to conduct NE treatment experiments.The mice uteri were extracted on day 5.Immunohistochemistry was used to detect the expression change of uterus macrophage membrane receptor CD14 and CD204.Results:Compared with the mice in the control group,after sympathetic blocking,the expression of CD14 at the endometrium,myometrium and perimetrium showed a significant high expression,and also exhibited a higher CD14^+macrophages distribution.The control group showed no expression of CD204,while the whole experimental group had the expression of CD204 except day 1 subgroup.And the quantity of expression increased over time.It reached the peak on day 9.After the injection of NE,the expression of CD14 and CD204 showed a significantly lower expression,and also exhibited CD14^+and CD204^+ macrophages distribution.And for different concentration groups,the medium and high groups down-regulated obviously.Conclusion:The sympathetic nerve and its transmitter can regulate the activity and function of macrophage by adjusting their amount and receptors of CD14 and CD204 to take part in the maintenance of uterine local immune status.
