摘  要:目的:观察米诺环素预处理对脊髓横断大鼠行为功能、不同时间胶质纤维酸性蛋白(GFAP)表达变化和超微结构的影响.方法:雄性Wistar大鼠随机分为3组,假手术组、模型组和米诺环素预处理组,采用改良Rivlin斜板实验、CBS评分对大鼠脊髓功能进行行为学评分.用免疫荧光分别检测3组大鼠在3、7、14 d GFAP的光密度值;透射电镜下观察各组大鼠脊髓损伤后21 d神经元和髓鞘的超微结构变化.结果:术后第3天模型组和米诺环素预处理组斜板临界度数及CBS评分无明显差别;术后第7、14、21d米诺环素预处理组斜板临界角度数均较模型组高.而米诺环素预处理组术后第7~21天CBS评分明显低于模型组.免疫荧光检查显示,米诺环素预处理组在3、7、14 d GFAP平均光密度值较模型组呈现出递减趋势.超微结构观察可见,米诺环素预处理组神经元和髓鞘的形态结构均优于模型组.结论:米诺环素预处理可有效改善脊髓横断大鼠不同时间GFAP蛋白的表达,抑制星形胶质细胞的活化增殖.促进神经功能的恢复,改善损伤脊髓组织神经元和髓鞘的超微结构.Objective:To observe the influences of minocycline pretreatment on behavioral function,GFAP at different time point expression and ultrastructure in rats with completely transected spinal cords.Methods:Sixty healthy adult male Wistar rats were randomly divided into sham operation group,model group and minocycline pretreated group(n = 20).Rats in minocycline group received minocycline injection intraperitoneally before operation(90 mg/kg).The function of spinal cord was evaluated via animal behavioral test,modified Rivilin loxotic plate test and CBS test.The immunofluorescence method was used to monitor the mean optical density of GFAP in each group at 3,7,14 and 21 days after the injury;The spinal cords were taken out for morphological studies under the transmission electronic microscope at 21 days after its injury.Results:Three days after spinal cord injury(SCI),rats in the model group and minocycline pretreated group showed no significant difference in the modified Rivilin loxotic plate test and CBS rating scale.But rats in the minocycline pretreated group had a significantly lower comblna behavior score in comparision with rats in the model group from 7 to 21 days after SCI(P<0.01);The mean optical density value(OD) of GFAP positive cells showed no obvious change in sham operation group at 3,7 and 14 days respectively tested by immunohistochemistry.The mean OD value of GFAP positive cells in injuried spinal cord increased significantly in model group,but showed a decreasing trend than that in minocycline pretreaed guoup at 3,7 and 14 days(P<0.01).Morphology of neurons and myelin sheath in minocycline pretreated group were better than that in model group based on the ultrastructure observation.Conclusion:Minocycline pretreatment can effectively improve the GFAP expression and inhibit the excessive proliferation of neuroglial cells in rats with transected spinal cord injury,promote the recovery of nerve function and improve the ultrastructure of injured spinal cord neuron and myelin sheath.
