鳖甲和三七对心肌成纤维细胞增殖影响的实验研究

摘　　要：目的:观察以鳖甲和三七为主要成分的复方鳖甲软肝片和氯沙坦对心肌成纤维细胞(cardiacfibroblasts,CFs)增殖的影响。方法:取生后1～3d的雄性SD仔鼠心脏原代培养心肌成纤维细胞、传代,将第3代心肌成纤维细随机分为3组:空白对照组、氯沙坦组、复方鳖甲软肝片组。并根据血清药理学理论,分别以蒸馏水、10mg/kg氯沙坦、0.7g/kg复方鳖甲软肝片喂养新西兰兔获得血清,制备相应培养液干预培养的心肌成纤维细胞。结果:鳖甲片和氯沙坦能明显抑制CFs增殖(空白对照组、氯沙坦组、鳖甲片组的A值分别为0.387±0.0539,0.181±0.044,0.212±0.038,F=5.805,P=0.016),并将细胞周期阻抑在G1期犤(74.32±1.76)%,(81.02±4.15)%,(81.62±3.83)%,F=7.041,P=0.009犦,处于分裂期(S期)的细胞比例减少犤(18.36±2.15)%,(13.18±3.26)%,(11.60±2.23)%,F=9.290,P=0.004犦,对CFs增殖和细胞周期影响的作用强度与氯沙坦相近(A值:95%CI:-0.180～1.118,P=0.662;G1和S期:95%CI:-5.306～4.106,P=0.768;95%CI:-1.995～5.515,P=0.355)。结论:复方鳖甲软肝片具有防治心肌纤维化的作用。AIM:To study the effects of biejia ruangan compound tablet, whose main compone nts are biejia and sanqi, and losartan on the proliferation of cardiac fibroblas ts (CFs). METHODS:CFs were cultured from male SD neonatal rat (1-3 days old) hearts by primary culture and passage, and those of the 3rd generation were randomly divid ed into three groups: control, losartan and biejia ruangan tablet group. Based o n the theory of serum-medicine, serums were obtained from New Zealand rabbits f ed by distilled water, losarton (10 mg/kg), biejia ruangan tablets (0.7 g/kg) re spectively, then corresponding culture media were made to intervene the culture of CFs. RESULTS: Biejia ruangan tablets and losarton could obviously inhibit the proli feration of CFs (the A values in the control, losartan and biejia tablet groups were 0.387±0.053 9, 0.181±0.044 and 0.212±0.038, respectively,F=5.805,P=0.016 ), and the cell cycle was controlled at G1 stage [(74.32±1.76)%,(81.02±4.1 5)%,(81.62±3.83)%,F=7.041,P=0.009].The proportion of cells at the divisi on stage (S stage) decreased [(18.36±2.15)%, (13.18±3.26)%, (11.60±2.23)%, F=9.290, P=0.004], and its intensity of effect on proliferation and CFs cell pr ogression of myofibroblasts was close to that of losartan(A value: 95%CI:-0.18 0-1.118,P=0.662;G1 and S stages:95%CI: -5.306-4.106,P=0.768;95%CI:-1.995- 5.515,P=0.355). CONCLUSION: Biejia ruangan compound tablet can prevent and cure cardiac fibros is.
