气质联用法监测尿中睾酮及其相关甾体激素

摘　　要：目的 :为了更好地监控运动员 ,保护运动员的身体健康 ,保证竞技体育的健康发展 ,降低滥用睾酮后假阴性出现的几率。方法 :利用GC/MS法对尿中睾酮及其相关甾体激素进行检测。结果 :在口服安雄后 ,尿中雄酮、苯胆烷醇酮、5α ,3α雄烷二醇、5 β ,3α雄烷二醇、表睾酮及睾酮的分泌均有增加 ,特别是雄酮、苯胆烷醇酮、5α ,3α雄烷二醇、5 β ,3α雄烷二醇及睾酮增加幅度较大 ,在用药后 2～ 6h内达到最高峰。在 6名志愿者的尿样中出现睾酮 /表睾酮 (T/E)比大于 6的仅有 2名 ,有的尿中的T/E即使与未用药前的对照尿样相比有所增加但比值也不大于 6 ,如按照国际奥林匹克委员会医学委员会将T/E >6∶1作为判断滥用外源性睾酮的依据 ,则会出现假阴性的结果 ;结论 :雄酮与表睾酮浓度比作为判定的依据之一 ,再结合睾酮表睾酮比要比单纯用T/E比来判定可减少假阴性的结果。AIM: Anabolic androgenic steroid(AAS) is one of primary performance-enhancing substances abused by athletes.To defeat doping controls,athletes have changed from the application of synthetic AAS to the application of endogenous androgens..Testosterone is one candidate of endogenous androgens abused in sports.The aims of this study are to forbid testosterone abusing to protect the health of athletes and assure the development of sports.METHOD: Detecting and monitoring testosterone and its relevant steroids in urine with GC/MS.RESULT: The concentration of androsterone,etiocholan-3α-ol-17-one,5α3α-androstane-3α,17β-diol,5β3α-androstane-3α-17β-diol,testosterone increased obviously after 80mg testosterone undecanoate taken orally.According to the rules of the International Olympic Committee,banned use of exogenous testosterone is estimated by a urinary ratio of testosterone/epitestosterone higher than 6 provided that there is no evidence of a physiological or pathological reason.In fact,the urinary ratio of T/E is less than 6 with testosterone undecanoate administration.There only the urines of 2 among 6 volunteers showed the ratio of T/E higher than 6.CONCLUSION: This study showed that both the urinary ratio androsterone/epitestosterone and T/E can help to enhance the detectable rate of positive urine and reduce miscarriage of justice.In addition,monitoring and controlling the urinary endogenous steroids of athletes with GC/MS is benefitial to forbiding testosterone abuse.
