摘  要:目的 :观察药物CPU86 0 17对左 甲状腺素 (L thy)所致的肥厚豚鼠心室肌细胞L 型钙电流的影响。方法 :ipL thy ,诱导豚鼠心肌肥厚 ,用全细胞膜片钳技术测定CPU86 0 17对心室肌细胞L 型钙电流强度和密度的影响。结果 :终浓度为 30 μmol/L的CPU86 0 17对正常豚鼠心室肌细胞L 型钙电流强度的抑制率达 74 2 1% ;肥厚组 ,终浓度为 30 0 ,10 0 ,3 3μmol/LCPU86 0 17均能明显抑制钙电流强度 ,抑制率分别为 6 7 94 % ,4 3 2 5 % ,2 7 0 7%。结论 :CPU86 0 17可抑制正常和肥厚豚鼠心室肌细胞膜上的L 型钙电流 ,可减轻肥厚所致的钙超载 ,有利于预防病变心脏的心律失常。AIM:To study the effects of CPU86017 on L -type calcium current (I_ Ca.L ) in single guinea pig hypertrophic ventricular myocytes induced by levothyroxine( L -thy). METHOD:After guinea pigs were ip L -thy 0.5 mg/kg.10 d,their heart was hypertrophic. Then we used whole-cell path-clamp recording technique to observe the effect of CPU86017 on I_ Ca.L in single ventricular myocytes. RESULT: The suppression by CPU86017 on I_ Ca.L in normal ventricular myocytes was 74.21% at 30.0 μmol/L.In hypertrophic animals the suppression by CPU86017 on I_ Ca.L was 67.94%,43.25% and 27.07% at 30.0,10.0,3.3 μmol/L,respectively. CONLUSION: CPU86017 was shown to inhibit I_ Ca.L in both normal and hypertrophic ventricular myocytes,so it can be used to prevent arrhythmia by decreasing calcium overload during hypertrophy.
