高效液相色谱法测定轻身减肥片中大黄素、大黄酚的含量

摘　　要：目的 　建立轻身减肥片中大黄素、大黄酚的含量测定方法。方法 　以 L ichrospher C1 8色谱柱 (4 .6mm× 2 5 0 mm,5 μm)为固定相 ,甲醇 -0 .1磷酸 (85∶ 15 )为流动相 ,柱温 3 0℃ ,检测波长为 2 5 4nm。 结果 　大黄素在 0 .0 3 45 6～ 0 .172 8μg范围内呈良好的线性关系 ,r=1.0 0 0 0 ,平均回收率为 99.95 % ,RSD=0 .42 % ;大黄酚在 0 .0 6912～ 0 .3 45 6μg范围内呈良好的线性关系 ,r=1.0 0 0 0 ,平均回收率为 99.19% ,RSD=0 .96%。结论　本法简便快速 ,结果准确 ,重现性好 ,可作为该制剂的含量测定方法。OBJECTIVE To establish a method for the determination of Emodin and Chrysophanol in Qingshen Jianfei Tablets by HPLC.METHODS Lichrospher C_ 18 column (4.6mm×250mm,5μm)was as stationary phase, Methanol-0.1% phosphoric acid(85∶15)as mobile phase, the temperature of column was set at 30℃ and the wavelength of detection was at 254nm.RESULTS The standard curve of Emodin was rectilinear within the range of 0.03456～0.1728μg,r=1.0000,the average recovery was 99.95%,RSD=0.42%;The standard curve of Chrysophanol was rectilinear within the range of 0.06912～0.3456μg,r=1.0000,the average recovery was 99.19%,RSD=0.96%.CONCLUSION This method is simple and quick, accurate and reproducible.
