摘 要:从河北、江西猪场中分离到3株猪繁殖与呼吸综合征病毒(HB-1(sh)/2002、HB-2(sh)/2002和JX-1/2002)。采用RT-PCR方法扩增其完整的ORF5基因片段,进行了序列测定,并与其它5个国内毒株的ORF5基因序列进行了比较和变异分析。所有这些毒株的ORF5基因均编码200个氨基酸,推导的氨基酸相似性在88.2%~99.0%之间。其中BJ-4、S1及J1间的相似性较高,在98%~99%之间;HB-1(sh)/2002,HB-2(sh)/2002,JX-1/2002及CH-1a间的相似性为92%~96%之间。根据ORF5基因核苷酸序列的遗传进化距离,这些毒株可分为两个亚群,BJ-4、S1和J1为一个亚群,与VR2332及疫苗毒株接近;HB-1(sh)/2002、HB-2(sh)/2002、JX-1/2002和CH-1a为另一个亚群。Three porcine reproductive and respiratory syndrome(PRRSV) isolates (HB-1(sh)/2002,HB-2(sh)/2002 and JX-1/2002) were obtained from Hebei and Jiangxi Provinces' pig farms. A RT-PCR method was used to amplify the complete ORF5 gene of the viruses. Variation analysis of ORF5 gene of the three isolates and other five published PRRSV isolates in China showed that all of these isolates belonged to American type, and percent identity of amino acid ranged from 88.2% to 99.0%. ORF5 gene among BJ-4, S1 and J1 had higher similarity, sharing 98%~99% identity of the deduced amino acid. HB-1(sh)/2002, HB-2(sh)/2002, JX-1/2002 and CH-1a showed 92%~96% identity among their ORF5 gene. Phylogenetic analysis suggested that these isolates could be divided into two subgroups according to the genetic distance. BJ-4, S1 and J1 belonged to one subgroup, which are more close to VR2332 and vaccine strains, whereas HB-1(sh)/2002, HB-2(sh)/2002, JX-1/2002 and CH-1a belonged to another subgroup.
