摘 要:目的 :研究富硒蘑菇 (seleniumenrichedmushroom ,SEM )对小鼠睾丸脂质过氧化和精子活力的影响并阐明其相关的自由基损伤机理。方法 :复制低硒小鼠模型 ,补充生理剂量范围的SEM、亚硒酸钠并设低硒和正常对照组 ,饲以低硒和普通小鼠饲料 ,历时 4 5d。测定睾丸硒 ,丙二醛 (MDA) ,一氧化氮含量和谷胱甘肽过氧化物酶 (GSH -Px)、一氧化氮合酶 (NOS)活性。用计算机辅助精子质量分析 (computerauxiliaryspermatozoaanalysis,CASA)技术测定小鼠精子活率、活力和运动方式等参数。结果 :SEM使小鼠睾丸硒含量增加 ,MDA和一氧化氮含量减少 ,NOS活性降低 ,谷胱甘肽过氧化物酶 (GSH -Px)活性增高 ,小鼠精子活率及运动能力加强。结论 :SEM具有提高含硒酶GSH -Px ,降低NOS活性 ,减轻睾丸自由基损害和增强小鼠精子活力的作用。Objective: To study the effect of selenium enriched mushroom (SEM) on sperm motility of mice and investigate its anti free radical damage mechanism. . Method: The male KunMing mice were used for experimentation, divided them into following groups: 1. SEM; administered with SEM in dosage 280 mg/(kg·d), 2. Selenium nitrate (Se); administered with selenium nitrate in dosage 112 μg/(kg·d), 3. Selenium deficient (Se-de) and 4. normal control (NC) ,the last two groups were administered with distilled water in same volume by oral injection. Groups SEM, Se, Se-de and NC were fed on selenium secareity diet or standard nomal diet of mice individually for 45 days. The testis concentration level of selenium, malonic aldehyde (MDA), nitric oxide (NO) and the activities of nitric oxide synthase(NOS), glutathione peraxidase (GSH-Px) of testis were measure. The computer assisted sperm analysis(CASA) was in use for evaluating sperm motive parameter. Result: All the mice, group of SEM : the sperm viability, motility of grade A+B and motive velocity and pattern parameters were increase significantly. The selenium content in the testis increased, but the testis index did not varied obviously, concentration level of MDA, NO and activities of NOS are decrease and GSH-Px is increase significantly. The group of Se possess all the above pharmacological effect. As compared with group of SEM and Se, they have same selenium content, the effect is enhancement trend to SEM. Conclusion: These data indicated that activities of GSH-Px and NOS, and the concentration level of NO, MDA and selenium could be changed after fed on SEM. It elevated sperm quality and suppressed the free radical damage in testis of mice.
