136例广东地区汉族人TGF-α/BamHI基因多态性分析

摘  要:目的 研究TGF -α基因BamHI限制性片段多态性在正常中国广东汉族人群的分布 ,计算各型的基因频率。方法 用盐提取法提取 136例广东地区正常汉族人白细胞的DNA ,用聚合酶链反应 -限制性片段长度多态性技术检测TGF -α基因BamHI多态性 ,并与其他人群做了比较。结果 TGF -α基因的BamHIA突变点A1(BamHI- )、A2 (BamHI+)等位基因频率分别为 6 %和 94 % ;A1A2和A2A2基因型频率分别为 12 5 %、87 5 %。与高加索人群比较无显著差异 (P >0 0 5 )。结论 该位点在中国广东汉族人群中具有多态性 ,与高加索人群不存在明显差异 ,为TGF -α基因BamHI多态性在广东正常汉族人群分布提供依据。Objective In order to study the distribution of TGF-α gene BamHI restriction fragment length polymorphism in normal guangdong Hans,and analyze the frequency of TGF-αgenotypes.Methods DNAs were extracted from the white blood cells of 136 healthy Guangdong Hans by salt-out method.Polymerase chain reaction-restriction fragment length polymorphism technique was used to detect TGF-α gene BamHI polymorphism.Current results were compared with the data on other ethnic group.Results Within the 136 individuals tested,the frequencies of the A1(BamHI -)and A2 (BamHI +)alleles of the BamHI A mutation site in TGF-α gene were found to be 6% and 94%,respectively.The observed frequencies of the A1A2 and A2A2 genotypes were found to be 12.5% and 87.5%,respectively.No significant difference was found between normal Guangdong Hans and normal Caucasian (P>0.05).Conclusion The polymorphism of TGF-α gene in Guangdong Hans exist,but no difference from that in Caucasian.
