日本血吸虫单克隆抗独特型抗体NP30保护性免疫力持续时间的研究

摘　　要：目的　确定日本血吸虫单克隆抗独特型抗体 NP30主动免疫诱导保护性免疫力的持续时间。方法　实验组昆明种小鼠腹腔注射 NP30 10μg/鼠 ,主动免疫 3次 ,每次间隔 2周 ,对照组腹腔注射生理盐水。末次免疫后 8、12、16、2 0、2 4周感染日本血吸虫尾蚴 (4 0± 1)条 ,尾蚴感染后 4 0 d行足垫试验。结果　 NP30末次免疫后 8、12、16周感染日本血吸虫尾蚴的减虫率为 2 1.4 3%～4 1.16 % ,分别与对照组相比差异有显著性 ;足垫试验结果显示 8、12周实验组与对照组差异有显著性。结论　 NP30主动免疫诱导的保护性免疫力持续约 4个月 ;Objective To determine the duration of the active protective immunity induced by the anti-idiotypic monoclonal antibody-NP30 of Schistosoma japonicum. Methods The mice were divided into two groups. The test group was intraperitioneally injected with NP30 10 μg for three times every two weeks, while the control group was injected with 0.9% sodium chloride solution intraperitoneally. Each mouse was infected with 40 cercariae of S. japonium in the abdominal skin at the eighth, twelfth, sixteenth, twentieth, twenty-forth week respectively after the third injection. The test of footpad in mice was conducted at the fortieth day after cercariae challenge. Results The reduction rate of adult worms ranged from 21.43% to 41.16% at the eighth, twelfth, sixteenth week after the third immunity of NP30, and when the test group compared with the control group there was a remarkable difference. The result of the footpad test showed that the test group compared with the control group had remarkable difference at the eighth, twelfth week after the third immunity. Conclusion The duration of the active protective immunity induced by the anti-idiotypic monoclonal antibody-NP30 of S. japonicum is about four months. The result of the footpad test is related to the cell immunity.
