骨髓瘤冻融物致敏树突状细胞诱导特异性细胞免疫反应

摘　　要：目的 研究多发性骨髓瘤(MM)患者肿瘤冻融物致敏的树突状细胞(DC)诱导T细胞对不同靶细胞产生的细胞毒作用。方法GM-CSF、IL-4和TNF-α体外联合诱导生成MM患者自身DC,患者骨髓瘤冻融物冲击致敏DC诱导自身淋巴细胞;MTT法检测致敏DC对不同靶细胞(患者骨髓瘤细胞、K562细胞株)的杀伤率及其与效靶比的关系。结果 当效靶比为20:1时,骨髓瘤细胞冻融物致敏的DC加IL-2诱导的自身T细胞对骨髓瘤细胞有明显的溶解作用,而它对K562细胞则无明显影响。结论 MM患者肿瘤冻融物致敏的DC能有效地诱导自身T细胞对其骨髓瘤细胞的特异性杀伤作用。Objective To explore the cytotoxic response of different antigens induced by dendritic cell ( DC ) pulsed with myeloma lysates in multiple myeloma ( MM) patients. Methods DC was obtained from bone marrow of mutiple myeloma patients and cultured with GM-CSF, IL-4 and TNF-α. Host lymphocytes were stimulated with DCs pulsed with myeloma lysates. MTT assay was performed in order to examine the specific lysing ability of cytotoxic T lymphocyte on different antigens (myeloma cells, K562 cell line). Results At the E/T ratio of 20:1 , autologous T lymphocytes induced by IL-2 and DCs pulsed with myeloma lysates had strong killing effect on myeloma cells, but had mild effect on K562 cells. Conclusion DCs pulsed with tumor antigens from multiple myeloma patients can effectively and specially induce autologous T cells to kill patients' myeloma cells.
