摘  要:目的 :检测不同发育时期脑组织神经干细胞巢蛋白 (nestin)的表达和发育变化规律 ,为分离、富集和增殖神经干细胞提供实验依据。方法 :将Wistar大鼠按胎龄和日龄分组 ,剥离脑组织 ,制成单细胞悬液 ,经间接荧光法标记nestin抗原 ,用流式细胞术检测并分析。结果 :E1 3至P90整个发育过程中都有nestin抗原的表达 ;从E1 3到E1 9表达逐渐升高 ,E1 9表达水平最高 ;出生后表达急剧下降 ,出生后 7d便接近于出生后 90d(成年鼠 )表达水平。结论 :胚胎发育近中期后 ,脑神经干细胞nestin表达水平随胎龄逐渐升高 ,E1 9达到高峰 ,出生后表达迅速下降 。Objective: To search for effective strategies to isolate, enrich and propagate homogeneous populations of neural stem cells (NSCs) in vitro, providing further information for the development mechanism of cerebral neuron. Methods: Wistar rats were divided into 9 groups according to different stages in the development: E13, E15, E17, E19, P1, P3, P5, P7, P90. One hemisphere of the brain was microsurgically obtained and made into single cell suspension. Nestin antigens of NSCs in the suspensions were combined with monoclonal antibodies of nestin; the compounds were then fixed with the second antibodies of IgG FITC, which were determined with flow cytometry. Results: Nestin antigens expressed during the investigation period with a trend of stable increase from E13 to E19, E19 at the summit, then drastically decreased and that of P7 was almost the same as that of P90. Conclusion: When embryo development approaches intermediate stages, the expression of nestin in the rat brain increases stably, E19 reaches the summit, and then decreases dramatically after born and reaches the level of adult within one week.
