Ⅱ型胶原移植修复关节软骨缺损过程中抗体水平的动态变化

摘　　要：目的建立检测Ⅱ型胶原抗体水平的间接法ELISA,研究猪Ⅱ型胶原移植修复兔关节软骨缺损过程中Ⅱ型胶原抗体水平的动态变化。方法取28只新西兰大白兔,随机分成2组。所有动物均在股骨外髁钻直径5mm、深3mm的软骨缺损区,深达软骨下骨,其中实验组在缺损区植入Ⅱ型胶原;另一组不植入Ⅱ型胶原作为对照。术前及术后每隔2周收集血清直至第12周,采用间接法ELISA检测血清中Ⅱ型胶原抗体。结果实验组Ⅱ型胶原抗体水平术后与术前比较均差异无显著性(P>0.05);而对照组术后第8周抗体水平显著高于术前(P<0.05);实验组与对照组同期比较,两者差异均无显著性。结论关节软骨缺损可引起机体自身免疫反应,产生一定水平的Ⅱ型胶原抗体。Ⅱ型胶原移植不但可以促进关节软骨缺损的修复,还可以降低机体的自身免疫反应,减少抗体的产生。Objective To establish the indirect ELISA to examine the levels of the antibody of typeⅡcollagen in serum and study the dynamic variations of the antibody after transplantation of typeⅡcollagen in the repair of articular cartilage defects in rabbits.Methods Twenty-eight mature New Zealand Rabbits were divided into two groups randomly.All rabbits were drilled a cartilage defect in femoral extramalleolus,which was5mm in diameter and3mm in depth and deep to the subchondral bone.Group1was filled with typeⅡcollagen.The other group was served as normal control without transplantion.Sera were collected be-fore and every two weeks after surgery up to the12week.Using indirect ELISA to detect the antibody of typeⅡcollagen in serum.Results Comparing preoperation with postoperation in group1,the antibody levels of typeⅡcollagen were not significantly different(P>0.05).In controls,there were significant difference between8th week postoperation and preoperation(P<0.05),whereas comparing group1with control group,there was no significant difference between them at the corresponding period.Conclusions Cartilage defect may cause au-to-immunity and lead to the levels of the antibody of typeⅡcollagen in serum rising.The transplantation of typeⅡcollagen can not only promote the repair of cartilage defect but also can reduce the reaction auto-im-munity and the production of the antibody of typeⅡcollagen in serum.
