摘 要:目的 :探讨冰冻切片的制作、HE染色和免疫组化的标准化实验方法 ,达到质量控制标准。方法 :详细介绍了冷冻切片的方法、影响因素、标准化与质量控制及其在临床快速病理诊断的 HE和免疫组织化学染色方法。结果 :经此方法制备的冷冻切片组织结构完整、细胞染色清晰、境界清楚、核质分明、对比度好 ,无刀痕、皱折、重叠、冰晶、切片平整、厚薄均匀一致等。结论 :可为临床快速病理诊断的 HE染色、脂肪黏液染色、酶组织化学、免疫组织化学、免疫荧光、核酸原位杂交以及原位Objective To explore a standard experimental method for making frozen sections and staining of hematoxylin eosin (H&E) and immunohistochemistry, the sections were to meet the quality control demand in clinic pathology tests. Methods This article described a series of sectioning, affective factors, standardization and quality control in making frozen slices and its staining methods of H&E and immunohistochemistry in clinic pathology diagnosis. Results The frozen sections by this method revealed markedly characteristic contrast, more detail and clear structure of tissues and cells, without the blade trace, fold, ice crystals and the phenomenon of thickness and thinness, and were suitable for microphotography. Conclusion This method can provide frozen sections superior in quality for clinic pathology diagnosis and research in stains of H&E, fat, mucus, enzyme histochemistry, immunohistochemistry, immunofluorescence, in situ hybridization and in situ PCR as well.
