大川芎丸对血管内皮细胞中血管内皮生长因子分泌水平的影响

摘　　要：目的:研究大川芎丸对体外培养人脐静脉内皮细胞中血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)分泌水平的影响,为临床用于缺血性疾病的治疗提供实验依据。方法:以体外培养人脐静脉内皮细胞为研究对象,分别在常氧和低氧培养条件下,采用中药血清药理学方法,在试验组培养液中加入含药血清,对照组加入空白血清,酶联免疫吸附法(ELISA)检测各组培养液中VEGF水平。结果:低氧培养条件下VEGF分泌水平犤(853.57±14.29)ng/L犦较常氧培养条件下犤(135.71±50.01)ng/L犦有明显升高(t'=30.862,P<0.001);试验组与对照组VEGF分泌水平在常氧犤(150.00±53.57)ng/L比(135.71±50.01)ng/L犦和低氧犤(839.29±13.57)ng/L比(853.57±14.29)ng/L犦两种培养条件下差异均无显著性意义(t=0.436,t'=0.079,P>0.05)。结论:低氧能有效促进体外培养人脐静脉内皮细胞VEGF的分泌,本试验选择浓度下的大川芎丸含药血清对VEGF的分泌无明显干预作用。AIM:To study the effect of da chuanxiong pill on the secretion level of vascular endothelial growth factor(VEGF) by human umbilical vein endothelial cells cultured in vitro,so as to provide experimental evidence for treatment of ischemic diseases. METHODS:According to the principle of seropharmacology of Chinese materia medica,human umbilical vein endothelial cells were cultured in vitro under the hypoxia and normoxia condition respectively,with contained herb serum added in culture media in experimental group and normal serum in control group,and the concentrations of VEGF in the culture medium were detected by enzyme linked immunosorbent assay(ELISA). RESULTS:The concentration of VEGF in hypoxia group[(853.57±14.29) ng/L] was significantly elevated as compared with that of normoxia group[(135.71±50.01) ng/L],and no significant difference of VEGF concentration was found between the experimental group and control group under both normoxia condition[(150.00±53.57) ng/L and(135.71±50.01) ng/L,respectively](t=0.279,P >0.05) and hypoxia condition[(839.29±13.57)ng/L and (853.57±14.29)ng/L, respectively](t=0.436,P >0.05). CONCLUSION:VEGF secretion by human umbilical vein endothelial cells cultured in vitro can be enhanced under hypoxia condition,and the serum containing da chuanxiong pill under the specified concentration has no significant interventional effect on the VEGF secretion.
