载脂蛋白(a)五核苷酸重复序列基因多态性与冠心病及血浆Lp(a)水平的关系

摘　　要：目的 :探讨载脂蛋白[apo(a)]五核苷酸重复序列基因多态性在冠心病患者中的分布状况及其与血浆Lp(a)水平的关系。方法 :对293例经冠脉造影确诊的冠心病患者及163例正常对照组进行apo(a)基因多态性频率分析及血浆Lp(a)水平的检测。PCR法扩增相应的apo(a)DNA片段 ,聚丙烯酰胺凝胶电泳分离扩增产物并确定其基因型。ELISA法测定血浆Lp(a)水平。结果 :共检出8种等位基因 ,19种基因型 ,常见的基因型为8/8、8/9、5/8、9/94种。2组中5/8基因型携带者的血浆Lp(a)水平均高于其他基因型携带者 (P<0.05) ;高Lp(a)组5/8基因型携带者多于低Lp(a)组 (P<0.05) ;且高Lp(a)组中冠心病患者所占比例明显高于低Lp(a)组(P<0.05)。结论 :5/8基因型携带者血浆Lp(a)水平升高 。Objective:To investigate the distribution of5pentanucleotide TTTTA repeat polymorphism of apo(a)gene in patients with coronary heart disease(CHD)and healthy controls,and to observe the relationship between polymorphism of apo(a)and serum Lp(a)levels.Methods:5pentanucleotide TTTTA repeat polyˉmorphism of apo(a)gene were analyzed in293patients with CHD and163healthy controls.The corresponding DNA fragments were amplified with PCR,and PCR products were separated by polyacrylamide gel elecˉtrophoresis.Serum Lp(a)levels were measured by ELISA.Results:Eight alleles and19genotypes were found totally.The common genotypes were8/8,8/9,5/8,and9/9,respectively.Serum Lp(a)level was higher in5/8genotype in both patients and controls(P<0.05).The higher distribution of5/8genotype in group with high Lp(a)and CHD group were observed(P<0.05).Conclusion:Serum Lp(a)was related to5/8genotype.Peoˉple with5/8genotype have higher risk for CHD.
