乳腺增生症增殖细胞核抗原表达的定量研究

摘　　要：目的 :探讨增殖细胞核抗原(PCNA)在乳腺增生症组织中表达的意义及其与乳腺癌的关系。方法 :应用免疫组化方法结合图象分析技术对根据Page标准分为 3级的 44例乳腺增生症(FCD)及 2 8例乳腺浸润性导管癌 (IDC)患者及 5例正常乳腺组织 (NMB)中的PCNA表达进行定量测定。结果 :PCNA含量按NMB、乳腺增生症Ⅰ级 (FCD1)、FCD2 、FCD3 及IDC的顺序递增 ,除FCD1与NMB组外 ,其余相邻组间差异均有统计学意义。结论 :1)PCNA含量增加是乳腺上皮不典型增生的一个重要特征 ;2 )PCNA的定量测定可以较为客观地评价乳腺上皮的增生程度及良、恶性。OBJECTIVE:To investigate the implication of proliferating cell nuclear antigen (PCNA) expression in the tissue of fibrocystic disease of breast (FCD) and its relation with mammary cancer.METHODS:By using the immunochemical method and image analysis technique (IAT),PCNA expression was quantitatively determined in 44 cases of FCD which were subdivided into 3 grades according to Page’s criterion, 28 case of invasive ductual carcinoma of breast (IDC) and 5 cases of human normal breast tissue (NMB).RESULTS:PCNA immunostaining was found in all cases.The mean PCNA level increased gradually in the order of NMB,FCD 1,FCD 2,FCD 3 and IDC.Except FCD 1 and NHB,the differences of mean PCNA level between the other close two groups were singnificant.CONCLUSIONS:1) In patients with FCD,over-expression of PCNA might be a maker of adenosis with atypical hyperplasia;2) PCNA mesurement might provide an objective assessment for quantification of malignant potential and hyperplasia of breast lesion.
