摘 要:酶法生产L-Met的后期分离效率较低一直是难以克服的问题。本文依据N-Ac-DL- Methionine的酶拆分液及其组分特有的性质,采用强酸型阳离子交换柱成功地将拆分液中各组分进行分组。分析了流出液的pH和L-Met浓度的变化规律和原因;讨论并确定了柱的交换容量与所加拆分液中钠的含量的最佳比值范围。本实验方法不仅可以获得L-Met,且兼得N-Ac-D-Met,二者的收率在小试中分别达到85%以上和75%左右。The N-acetyl-D-methionine, NaAc and small amount of N-acetyl-L-methionine in the solution attained by enzymatic deacylation of N-acetyl-DL-methionine will drastically affect the yield of the crystallization of L-methionine. In present research, different components were successfully grouped through a H type ion-exchange column, based on the special properties of this solution before concentration in order to increase the yield. The separation conditions have been optimized by investigating the effects of pH values and concentration of L-Met and the scope of optimum ratio between the content of sodium cation in the solution and the ion-exchange capacity on the separation efficiency. The results indicate that under these optimized conditions the yields of L-methionine and N-acetyl-D-methionine, reached 85% and 75%, respectively.
