摘　　要：介绍了一种改进的免疫双向扩散方法及其在重组呼吸道合胞病毒(RSV)疫苗研制中的用途。本法在琼脂凝胶中附加0.1μg/mL左右的溴酚蓝,使凝胶呈淡蓝色,与抗原-抗体复合物产生的白色沉淀线形成较大的反差,可显示出比较微弱的沉淀线,因而改善了免疫双向扩散法的灵敏度。用这种改进的方法检测RSV抗血清滴度、筛查RSV阳性小鼠以及检测重组RSV蛋白疫苗的抗原反应性,均取得了较传统方法更佳的效果。A modified double diffusion method and its applications in the studies of recombinant protein vaccine against RSV were described in this paper.About 0.1μg /mL of bromophenol blue was added in the agarose gel in present method,which makes the gel has faint blue or clear blue background.So,a greater contrast between the gel background and precipitin bands of antibody-antigen complexes is obtained.When viewed the bands against an appropriate light,even faint bands are visible.As a result,the sensitivity of double diffusion is improved.Better results were produced by this method than the traditional one when it was used in the titer test of rabbit serum against RSV,checking RSV antibody in the blood of mice and examining the antigen reactivity of recombinant RSV proteins.
