摘  要:以转抑制衰老有关的异戊烯基转移酶 (IPT)基因株系GC 1、携带抗白叶枯病基因Xa2 3的“CBB2 3”和抗稻瘟病水稻品种“合系 15号”为供体 ,采用分子标记辅助选择与生物学鉴定相结合的方法 ,将IPT基因与Xa2 3及抗稻瘟病基因进行聚合。在 3个复交组合中获得了 17株聚合IPT基因与Xa2 3基因的植株 ,用这些植株与两系杂交稻亲本 9311、E32、培矮 6 4S及W9834S进行杂交和回交 ,经PCR分子检测和抗白叶枯病、抗稻瘟病鉴定和细胞分裂素含量的测定 ,最终在 4个BC1回交组合“(9311/ / /合系 15 /CBB2 3∥GC 1)× 9311”、“(E32 / / /合系15 /CBB2 3∥GC 1)×E32”、“(培矮 6 4S/ / /合系 15 /CBB2 3∥GC 1)×培矮 6 4S”和“(GC 1/CBB2 3/ /W 9834S/合系 15 )×W 98 34S”中获得了 17株携带IPT基因与Xa2 3基因的BC1F1植株 ,这些植株对来自北方稻区 2 1个稻瘟病菌系全部表现为抗。携带IPT基因的抗病植株再与杂交稻亲本进行回交 ,在 2个BC2 回交组合“[(9311///合系 15 /CBB2 3∥GC 1)× 9311]× 9311”和“[(E32 ///合系15 /CBB2 3∥GC 1)×E32 ]×E32”中获得了 7株携带IPT基因与Xa2 3基因的植株 ,这些植株再经过 1~ 2代回交和自交 。Transgenic lines (GC-1) carrying a senescence-inhibition cheimeric gene,IPT(isopentenyl transferase) gene,CBB23,a isogenic lines carrying Xa23 gene for resistance to bacterial blight,and Hexi15,a commercial cultivar showing high resistance to blast disease,were used as donors to pyramid IPT gene and Xa23 by marker-assisted selection (MAS).Seventeen BC 1F 1 plants pyramiding Xa23 gene and IPT genes were obtained from three multi-cross combinations.Then,the plants carrying Xa23 and IPT genes were crossed with parental lines of two-line hybrid rice,such as 9311,E32,Pei’ai 64S and W9834S.The progenies were backcrossed the acceptor parents.A total of 17 plants carrying Xa23 and IPT genes were detected by PCR,disease resistance (identification) and analysis of CTK contents of in the four combinations of “(9311///Hexi15/CBB23∥GC-1)×9311”,“(E32///Hexi15/CBB23∥GC-1)×E32”,“(Pei’ai 64S///Hexi15/CBB23∥GC-1)×Pei’ai 64S” and “(GC-1/CBB23∥W9834S/Hexi15)×W9834S”.These plants showed resistance to blast disease by inoculating test using 21 the lines of Pyricularia grisea from Northern China.Six plants of BC 2F 1 pyramiding Xa23 and IPT genes were further obtained in the combinations of “[(9311///Hexi15/CBB23∥GC-1)×9311]×9311”,“[(E32///Hexi15/CBB23∥GC-1)×E32]×E32”.After backcrossed and self-crossed 1~2nd,the plants pyramiding Xa23 and IPT genes can be used in the program of hybrid rice breeding.
