摘  要:目的 探讨化学因子对辐射损伤小鼠脾淋巴细胞增殖的影响。方法 采用3H -TdR掺入法观察小鼠脾淋巴细胞增殖的变化。结果 与刀豆球蛋白A(ConcanavalinA ,ConA)同时作用于小鼠脾淋巴细胞的 10 - 6 mol L和10 - 5mol L的H2 O2 对ConA刺激的小鼠脾淋巴细胞增殖有明显的促进作用 ,H2 O2 在ConA作用于小鼠脾淋巴细胞 5 6h加入 ,H2 O2 对小鼠脾淋巴细胞增殖表现为抑制效应。当环磷酰胺 (cyclophosphamide ,CP)浓度≥ 10 - 4mol L和≥ 10 - 3mol L时 ,CP对脂多糖 (lipopolysaccharide ,LPS)和ConA刺激的小鼠脾淋巴细胞增殖分别表现为明显的抑制作用。当H2 O2浓度≥ 10 - 6 mol L和CP浓度≥ 10 - 3mol L时 ,H2 O2 和CP对辐射损伤的小鼠脾淋巴细胞增殖具有明显的抑制作用。结论 低浓度H2 O2 可以诱导小鼠脾淋巴细胞增殖的适应性反应 ,而H2 O2Objective To study the effect of chemical factors on the proliferation of splenic lymphocytes in irradiated mice. Methods To investigate the proliferation of splenic lymphocytes using 3H-TdR incorporation.Results The hydrogen Peroxide(10 -6mol/L and 10 -5mol/L) increased significantly the proliferation of splenic lymphocytes induced by Con?A (concanavalin A) when hydrogen Peroxide and Con?A were simultaneously added into splenocytes in mice. The proliferation of splenic lymphocytes was decreased significantly to add the hydrogen Peroxide at 56?h later. It was lower than control induced by LPS (lipopolysaccharide) and Con?A When the concentration of CP (cyclophosphamide) was ≥10 -4mol/L(LPS group) and ≥10 -3mol/L(Con?A group). It was also decreased significantly when the concentration of hydrogen Peroxide was ≥10 -6mol/L and the concentration of CP was ≥10 -3mol/L in irradiated mice. Conclusion Low dose hydrogen Peroxide may induce the adaptive response and hydrogen Peroxide and CP could not induce the resistance to irradiation on the proliferation of splenic lymphocytes in mice.
