摘  要:目的观察三七总皂甙对兔VSMC生长增殖、细胞周期和NF-κB活性的影响 ,以探讨其药理机理。方法胎牛血清刺激体外培养的兔VSMC使其快速增殖生长 ,加入不同浓度的三七总皂甙作用一定时间后 ,通过MTT法测定三七总皂甙对VSMC生长的抑制率、流式细胞技术测定细胞周期和NF -κB活性的影响。结果三七总皂甙能明显抑制胎牛血清刺激的VSMC的增殖 (IC50 为14.8μg/ml)。三七总皂甙还以浓度依赖方式减少S期细胞 ,阻滞细胞于G0/G1 期 ,并下调NF-κB活性 ,与对照组相比具有显著差异 (P<0.01)。结论三七总皂甙能显著抑制VSMC的增殖生长 。Objective To investigate the effect of total panax notoginseng saponins(tPNS)on the proliferaˉtion,cell cycle and NF-κB activity of rabbit vascular smooth muscle cell(VSMC)in order to find out the mechanism of its action.Methods By techniques of cell culture in vitro,VSMC stimulated by fetal bovine serum were treated by tPNS in different concentration for a certain period,then the inhibitory rate of cell proliferation was meaˉsured by MTT method,cell cycle distribution and NF-κB activity of the cell were measured by Flow-Cytometry respectively.Results tPNS could obviously inhibit the proliferation of VSMC stimulated by fetal bovine serum(IC 50 value was14.8μg/ml).tPNS also could decrease S phase cells,increase G 0 /G 1 phase cells and down-regulate NF-κB activity of VSMC by concentration-dependent manner,and the results were significant compared with the control(P<0.01).Conclusions tPNS could obviously inhibit the proliferation of VSMC,and regulating NF-κB activity of VSMC may be one of the mechanisms of tPNS'preventive atherosclerosis.
