和脉合剂中葛根素的测定

摘　　要：对和脉合剂中所含的葛根素进行定性定量测定。采用薄层层析法 (TLC法 )对和脉合剂中的葛根素进行定性鉴定 ,并应用Agilent 110 0seriesHPLC色谱仪 (配DAD检测器 )对葛根素进行含量测定 ,色谱柱为ZOR BAX。甲醇 :水 (2 8:72v/v)作流动相 ,用外标法进行定量测定。薄层层析结果显示 ,供试品色谱中 ,在与对照品溶液及对照药材溶液的相应位置上显相同颜色的斑点 ,该法简便 ,重现性好。定量测定结果显示平均加样回收率为98 2 %。和脉合剂中葛根素的含量为 0 0 5 % ,RSD为 1 0 6 % (n =3)。此方法准确、敏感、可靠 。To identify and determine the contents of puerarin in 'Hemai Mixture', the puerarin was identified by TLC method, and the content of puerarin by HPLC. Chromatographic conditions: Agilent 1100 series HPLC with DAD detector, ZORBAX column, methanol and water (28:72 v/v) as mobile phase. The determination was conducted by the externat standard at 250 nm. Results: The linearities were fairly good; the average recovery was 98.2%; the content of puerarin was 0.05% with RSD of 1.06% (n=3). It is proved that the methods are accurate, sensitive and reliable, and are suitable for the analysis of 'Hemai Mixture'.
