疏肝活血中药复方对B16小鼠黑素瘤细胞黑素合成的影响

摘　　要：目的探讨疏肝活血中药复方治疗黄褐斑的作用机制。方法以系列浓度的疏肝活血中药复方水煎剂作用于体外培养的B16小鼠黑素瘤细胞,测定细胞酪氨酸活性、黑素含量和细胞增殖率,琼脂糖凝胶电泳检测细胞凋亡。结果疏肝活血中药复方水煎剂能剂量依赖性抑制体外培养的B16小鼠瘤细胞酪氨酸活性和黑素合成,抑制细胞增殖,但无细胞凋亡发生。结论疏肝活血中药复方可直接作用于黑素细胞,抑制细胞黑素合成,但无细胞毒作用,可用于治疗黄褐斑。Objective Clinical studies have demonstrated that oral improves hyperpigmented lesions of melasm. The basic mechanisms underlying this effects are not known. In this study, we studied the effects of the deoction for soothing liver and activating blood on the melanogenesis in B16 melanoma cells to elucidate this problem. Methods The deoction for soothing liver and activating blood was incubated with B16 melanoma cells in vivo at serial concentrations, and then melanin content and tyrosinase activity were measured. In addition, the effects on induction of apoptosis in cells were detected by gel electrophoresis method. Results The deoction for soothing liver and activating blood decreased the activity of the enzyme tyrosinase and the level of melanin content in a dose-dependent manner, and typical DNA ladder was not found during gel electrophoresis. Conclusion The deoction for soothing liver and activating blood inhibit melanogenesis directly in B16 melanoma cells, which may explain by part the mechanism of skin-whitening role of it in melasm.
