摘 要:目的 通过emm基因序列分析对A群链球菌 (GAS)进行分型 ,探讨T型与emm型的对应关系。方法 用引物以PCR法扩增GASM蛋白的N末端并进行测序 ,确定GAS的emm型 ,并与T分型对照比较。结果 (1) 10 4株菌株中 ,emm1所占比例最大 ,为 2 9.8% ,然后依次是emm18(9.6 % ) ,emm12 (7.7% ) ,和emm6 9(5 .8% )等。 (2 )T1、T12和T14 4 9与emm基因对应关系好 ,T116株全部对应emm1型 (10 0 % ) ,T12对应emm12 ,符合率 83.3% ,T14 4 92株均对应emm4 9型 ;其它菌型对应关系差 ;同一T型可对应数个emm基因型 ,同一emm基因型可对应不同的T型。 (3)发现 1株新的emm型菌株 ,序列已递交GenBank ,注册号为AY3472 5 9。结论 emm基因分型方法具有准确、快速、分辨力高的优点 ;我国部分地区A群链球菌emm基因分型主导型为emm1、18、12、6 9、110等菌型。Objective To examine the type distribution of pathogenic group A streptococcal(GAS) strains in China and the corresponding relation to the T-agglutination type and M protein gene(emm) type. Methods emm gene was sequenced for the 104 isolates of Streptococcus pyogenes collected from school children in China from 1988-1994 and compared with their T type. Results (1) Among the 104 strains, emm1 was most dominant type as for 29.8%, followed by emm18(9.6%), emm12(7.7%) and emm69 (5.8%). (2) Some T type exhibited a good percentage of agreement with the emm sequence type such as T1, T12 and T14/49. 16 T1 strains all agreed with emm1(100%), 5 of 6 T12 strains permitted with emm12 (83.3%), and 2 T14/49 strains all matched emm 49; the other strains had varied T/emm pattern associations such as T13, T27. In addition, difference emm gene type matched to the same T type, and the same emm type had several varies T type. (3) A new sequence type that did not show homology to any published emm type in GenBank was characterized and submitted to GenBank(AY347259). Conclusion The emm typing method dose not require many kinds of M typing antisera, and it can characterize the M protein accurately and rapidly by sequencing the emm-specific PCR products. Therefore, this method is an useful tool for typing Streptococcus pyogenes isolates. The study show the predominating emm sequence type of GAS in some regions of China is emm1, emm18, emm12, emm69 and emm110, this result provides an useful guideline to develop the M-type specific vaccine for RF in a specific area of endemicity.
