摘　　要：目的 :观察不同浓度拓扑替康 (TPT)对人卵巢癌细胞系SKOV3中Survivin表达的影响。方法 :采用免疫组化和RT PCR方法 ,检测体外不同浓度TPT(0 .2 4 4 μg/L ,3.90 6 μg/L ,6 2 .5 μg/L ,1mg/L ,1 6mg/L ,1 2 8mg/L)作用于SKOV348h后SurvivinmRNA及蛋白的表达。结果 :随TPT浓度的增加 ,SKOV3细胞中SurvivinmRNA及蛋白的表达明显减少 ,呈正性剂量效应关系 (P <0 .0 5 )。结论 :TPT能够抑制卵巢癌细胞SKOV3中SurvivinmRNA和蛋白的表达。Aim: To investigate the effect of topotecan on the expression of Survivin in ovarian cancer cell. Methods:SKOV3 cells were cultured with different concentration of topotecan(0.244 μg/L,3.906 μg/L,62.5 μg/L,1 mg/L,16 mg/L,128 mg/L),then the Survivin protein and mRNA expression of SKOV3 were investigated by immunohistochemistry and RT PCR, respectively. Results:Under the effect of topotecan, expression of Survivin mRNA and protein significantly decreased, which showed a dose effect response. Compared with control group, the difference was significant ( P <0.05). As time and concentration of TPT increased, the expression of Survivin mRNA and protein were inhibited distinctly. Conclusion: TPT may inhibit expression of Survivin mRNA and protein in SKOV3 cells.
