摘  要:目的:研究石菖蒲挥发油中β-细辛醚在家兔及小鼠体内的药动学。方法:采用毛细管气相色谱法测定β-细辛醚的血药浓度,色谱柱:HP-35;程序升温:起始温度为30℃,6℃·min-1升至220℃,保持5 min;进样口温度250℃;检测器温度320℃;进样量2.0μL;不分流进样;载气:氦气;柱头压:5.5×104 Pa;内标物:α-萘酚。结果:建立了用气相色谱法测定β-细辛醚血药浓度的方法,线性范围0.04-40mg·L-1,全血中最低检测浓度0.04mg.L-1。结论:家兔或小鼠口服石菖蒲挥发油后,β-细辛醚在体内的药时过程均为线性动力学过程,符合一级吸收二室模型,而小鼠tmax,T1/2α以及T1/2β均提前,绝对生物利用度明显提高。AIM: To study the pharmacokinetics of β-asarone in volatile oil from Acorus Tatarinowu Schott in rabbit and in mice. METHODS: A capillary gas chromatography(GC) was applied to determine the concentration of β-asarone in blood. The GC system include: HP-35 column. Helium as carrier gas, oven temperature started at 30℃, then raised to 220℃ by a step of 6℃· min-1, stayed for 5 min, injector temperature: 250℃, detector temperature: 320℃. Injection volume was 2.0 μL with splitless inject mode. Methanol was used as solvent and a-naphthol as interior standard substance. RESULTS: A capillary GC method was established for determination of β-asarone in rabbit blood. The linear equation: Y=0.019c + 1.755×10-3(n= 6, r = 0.9990) was obtained in the range of 0.04-40 mgμL-1( Y: ratio of the peak area of β-asarone and of a-naphthol, c: concentration of β-asarone in blood) . The limit of quantification 0.04 mg·L-1 was achieved. CONCLUSION: After oral administration of volatile oil from Acorus Tatarinowii Schott, the inner body process of β-asarone in rabbit accords with two-compartment model with 1st order absorption. The T1/2β and T1/2α are 18.3 min and 4.5 min respectively. And in mice, the inner body process of β-asarone accorded with two-compartment model with 1st order absorption too. The tmax,T1/2β and T1/2β are 10 min, 6.5 min and 93.6 min respectively.
